【摘要】：目的 :從江蘇省2012年4月1例菲律賓輸入的疑似登革熱患者急性期血清中分離病毒并分析其分子生物學特征,以找出病原學證據(jù)。方法:收集患者血清樣本,采用膠體金法檢測登革病毒的Ig M、Ig G抗體;用C6/36細胞分離病毒,對細胞病變的標本進行RT-PCR及熒光定量PCR檢測登革病毒RNA和鑒定型別;采用高通量測序技術(shù)對該分離株進行編碼區(qū)基因測序,用MEGA6.0軟件對其進行序列比對和進化分析。結(jié)果:從患者血清樣本中檢測到登革病毒的Ig M抗體,分離到的毒株經(jīng)鑒定為登革病毒1型,與美國Haw O3663毒株的核苷酸同源性98%。結(jié)論:該病例是由登革病毒1型引起的輸入性病例。
[Abstract]:Objective: to isolate the virus from the serum of a suspected dengue fever patient imported from the Philippines in Jiangsu Province in April 2012 and analyze its molecular biological characteristics in order to find out the etiological evidence. The immunoglobulin G antibody of dengue virus was detected by colloidal gold assay, the virus was isolated by C6 / 36 cells, and the RNA and type of dengue virus were detected by RT-PCR and fluorescence quantitative PCR in cytopathic samples. The coding region gene of the isolate was sequenced by high-throughput sequencing technique. Sequence alignment and evolutionary analysis were performed by MEGA6.0 software. Results: the IgG antibody of dengue virus was detected from patient's serum samples. The isolated strain was identified as dengue virus type 1, which was homologous to the nucleotide sequence of Haw O3663 strain in the United States. Conclusion: this case is an imported case caused by dengue virus type 1.
