【摘要】:目的為了快速、準確、便捷的檢測和鑒定布魯氏菌,本研究擬建立鑒定布魯氏菌屬的MALDI-TOF-MS方法,利用現(xiàn)場分離菌株對該方法進行特異性和敏感性評價。方法收集布魯氏菌標準菌株和地方流行株,應用MALDI-TOF-MS采集圖譜,獲取獨特的蛋白質指紋圖譜,匯總成標準圖譜,建立布魯氏菌鑒定數(shù)據(jù)庫。并用39株布魯氏菌菌株,對建立的數(shù)據(jù)庫進行驗證。結果經(jīng)數(shù)據(jù)庫鑒定,39株布魯氏菌菌株與數(shù)據(jù)庫中布魯氏菌的匹配分數(shù)全部大于2.300,均報告為布魯氏菌,表明鑒定結果的可信度很高。聚類分型結果表明,在蛋白質水平,MALDI-TOF-MS將測試的布魯氏菌分成3大類。結論MALDI-TOF-MS對布魯氏菌進行鑒定,具有快速、準確、靈敏等優(yōu)點,可以實現(xiàn)對布魯氏菌屬的準確鑒定,對布魯氏菌病的臨床診斷具有較高的使用價值。
[Abstract]:Objective in order to detect and identify brucella quickly, accurately and conveniently, a MALDI-TOF-MS method for identification of Brucella was established in this study. The specificity and sensitivity of the method were evaluated by using field isolated strains. Methods the standard strains and endemic strains of Brucella were collected, and the unique protein fingerprints were obtained by using MALDI-TOF-MS. A database of brucella identification was established and 39 strains of brucella were used. Results 39 strains of Brucella and Brucella in the database were all identified as brucella, all of which were reported as brucella. The results of cluster typing showed that the tested brucella could be classified into 3 groups at protein level. Conclusion the identification of Brucella by MALDI-TOF-MS has the advantages of rapidity, accuracy, sensitivity and so on. It can be used to identify brucellosis accurately and has high value for clinical diagnosis of brucellosis.
