【摘要】：目的篩選結核分枝桿菌(tuberculosismycobacterium,MTB)與非結核分枝桿菌(non-tuberculosismycobacterium,NTM)感染肺部結核病患者血漿中差異表達microRNAs(miRNAs)分子。方法收集唐山市第四醫(yī)院與石家莊第五醫(yī)院2013.10-2014.10期間確診的NTM感染肺結核患者19例作為NTM組,MTB感染肺結核患者37例作為MTB組;抽取EDTA抗凝血并分離血漿。從兩組中分別取6例研究樣本,采用miRNAs芯片篩選差異表達miRNAs,其余樣本采用RT-q PCR技術進行后期驗證。結果與MTB組相比,NTM組差異表達顯著的miRNA共11個,其中上調(diào)的有8個,分別為miR-26a,miR-24,miR-222,miR-191,miR-155,miR-126,miR-122和let-7b;下調(diào)的有3個,分別為miR-767-3p、miR-1283、miR-1281。分別選擇上調(diào)明顯的3個上調(diào)和下調(diào)的miRNA進行RT-q PCR驗證(has-let-7b、has-miR-155、has-miR-26a、has-miR-1281、has-miR-1283、has-miR-767),驗證結果與芯片篩選結果具有較好的一致性。結論 MTB與NTM感染肺部結核病患者血漿中存在差異表達miRNAs分子。
[Abstract]:Objective to screen the differential expression of microRNAsmiRNAs-molecules in plasma of patients infected with pulmonary tuberculosis by Mycobacterium tuberculosismycobacterium tuberculosis (MTB) and non-tuberculous Mycobacterium tuberculosis (NTM). Methods the pulmonary nodule of NTM infection diagnosed between the fourth Hospital of Tangshan City and the Fifth Hospital of Shijiazhuang between March 10 and October 2014 was collected. Nuclear patients were treated as NTM group (n = 37), pulmonary tuberculosis group (n = 37) and MTB group (n = 37). EDTA anticoagulant was extracted and plasma was isolated. Six samples were collected from the two groups, and the differential expression of miRNASS was screened by miRNAs microarray, and the other samples were verified by RT-q PCR technique. Results compared with MTB group, there were 11 miRNA samples with significant differentially expressed miRNA. Eight of them, miR-26afmiR-24, miR-222, miR-191, miR-155, miR-126, miR-122 and let-7b, were down-regulated, and three were down-regulated. Three up-regulated and down-regulated miRNA were selected to verify the expression of miRNAs molecules in the plasma of patients with pulmonary tuberculosis infected with MTB and NTM respectively by RT-q PCR verification of has-let-7bsi-has-miR-155has-miR-1281Hhas-miR-1283has-miR-7671.Conclusion there is a difference in expression of miRNAs molecules between MTB and NTM infected patients with pulmonary tuberculosis.
