福建省2000-2015年Ⅱ型脊髓灰質(zhì)炎疫苗相關(guān)株病毒變異特征分析

【摘要】:目的了解福建省II型脊髓灰質(zhì)炎(脊灰)疫苗相關(guān)株病毒(VAPV2)變異特征,為證實(shí)阻斷VAPV2傳播提供依據(jù)。方法采用RT-PCR擴(kuò)增VAPV的VP1全長(zhǎng)和部分3D區(qū)基因序列,統(tǒng)計(jì)分析病毒核苷酸(nt)和氨基酸(aa)變異情況。結(jié)果 2000-2015年福建省從5 497份糞便標(biāo)本中分離到219株脊灰病毒,1株為疫苗衍生脊灰病毒(VDPV),218株為VAPV,其中VAPV2占37.16%。與Sabin2株相比,47株VAPV2中VP1變異位點(diǎn)5個(gè)2株(4.26%),≤2個(gè)36株(76.60%);共涉及45個(gè)位點(diǎn),其中nt2909為突變熱點(diǎn),導(dǎo)致VP1的aa143位點(diǎn)從非極性疏水性轉(zhuǎn)換成極性親水性。13株VAPV在3D區(qū)出現(xiàn)重組。結(jié)論福建省大部分VAPV2變異少,VP1的aa143位點(diǎn)為突變熱點(diǎn)。
[Abstract]:Objective to investigate the variation characteristics of poliomyelitis type II virus (VAPV2) in Fujian Province, and to provide evidence for blocking the transmission of VAPV2. Methods RT-PCR was used to amplify the full length and partial 3D region gene sequence of VAPV. Results from 2000 to 2015, 219 strains of poliovirus were isolated from 5 497 fecal specimens in Fujian Province. One strain of poliovirus was vaccine-derived poliovirus, and 218 strains were vaccine-derived poliovirus, of which VAPV2 accounted for 37.16%.
