TLR2和TLR4在結(jié)核分枝桿菌誘導(dǎo)腎小管上皮細(xì)胞損傷中的作用

【摘要】:目的:探討Toll樣受體2(TLR2)和Toll樣受體4(TLR4)在牛型結(jié)核桿菌卡介苗(BCG)誘導(dǎo)人腎小管上皮細(xì)胞損傷中的作用。方法:用BCG刺激HK-2細(xì)胞,通過(guò)熒光定量PCR或Western blot方法分別檢測(cè)TLR2、TLR4、轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)、趨化因子CX3CL1、E-鈣粘蛋白(E-cad)和α-平滑肌肌動(dòng)蛋白(α-SMA)的表達(dá)。將TLR2抗體以及TLR4拮抗劑CLI-90分別與BCG共刺激HK-2細(xì)胞,通過(guò)熒光定量PCR或Western blot方法分別檢測(cè)TGF-β1、CX3CL1、E-cad和α-SMA的表達(dá)水平。結(jié)果:BCG刺激HK-2細(xì)胞可誘導(dǎo)TLR2、TLR4、TGF-β1、CX3CL1、α-SMA表達(dá)上調(diào)及E-cad表達(dá)下調(diào)。其中TLR2和TLR4的表達(dá)呈時(shí)間依賴(lài)性和劑量依賴(lài)性,TGF-β1、CX3CL1、α-SMA和E-cad表達(dá)則表現(xiàn)為時(shí)間依賴(lài)性。TLR2抗體和TLR4拮抗劑可部分抑制BCG引起的TGF-β1、CX3CL1、E-cad和α-SMA表達(dá)的變化。結(jié)論:BCG可通過(guò)激活TLR2和TLR4引起TGF-β1、CX3CL1、α-SMA表達(dá)上調(diào)及E-cad表達(dá)的下調(diào),提示TLR2和TLR4介導(dǎo)的信號(hào)通路在結(jié)核分枝桿菌致腎小管上皮細(xì)胞損傷中發(fā)揮重要作用。
[Abstract]:Objective: to investigate the application of Toll like receptor 2 (TLR2) and Toll like receptor 4 (TLR4) in BCG vaccine of Mycobacterium bovis. Methods: BCG was used to stimulate HK-2 cells. TLR2, TLR4, TGF- 尾 1 and CX3CL1 were detected by fluorescence quantitative PCR or Western blot, respectively. E-cadA and 偽 -SMAA. TLR2 antibody and TLR4 antagonist CLI-90 were co-stimulated with BCG in HK-2 cells. TGF- 尾 1 CX3CL1 was detected by fluorescence quantitative PCR or Western blot. Results: TLR2TLR4TGF- 尾 1 and CX3CL1 could be induced in HK-2 cells stimulated by TLR2TLR4 and 偽 -SMA. The expression of 偽 -SMA was up-regulated and the expression of E-cad was down-regulated. The expressions of TLR2 and TLR4 were time-dependent and dose-dependent. The expression of 偽 -SMA and E-cad was time-dependent. TLR2 antibody and TLR4 antagonist could partially inhibit TGF- 尾 1 CX3CL1 induced by BCG. Conclusion the expression of TGF- 尾 1 and CX3CL1 can be induced by activation of TLR2 and TLR4. The up-regulation of 偽 -SMA and down-regulation of E-cad suggest that TLR2 and TLR4 mediated signaling pathway play an important role in renal tubular epithelial cell injury induced by Mycobacterium tuberculosis.
