熊去氧膽酸對膽汁酸腸肝循環(huán)動力學(xué)的作用研究

背景及目的近年來,科學(xué)家發(fā)現(xiàn)膽汁酸(bile acids,BAs)不僅可以作為乳化劑促進(jìn)脂類營養(yǎng)物質(zhì)、脂溶性維生素的吸收,還可以作為信號分子通過調(diào)節(jié)多種受體參與糖、脂、能量代謝并維持自身平衡。膽汁酸的核受體之一法尼酯X受體(farnesoid x receptor,FXR)在膽汁酸的腸肝循環(huán)以及維持膽汁酸穩(wěn)態(tài)方面有重要調(diào)節(jié)作用,然而不同膽汁酸對FXR的作用以及對膽汁酸腸肝循環(huán)動力學(xué)的影響目前尚未有明確的結(jié)論。熊去氧膽酸(ursodeoxycholic acid,UDCA)作為膽汁酸的一種,具有利膽、抗炎、細(xì)胞保護(hù)的特性,是被美國食品藥品監(jiān)督管理局(food and drug administration,FDA)認(rèn)可的治療原發(fā)性膽汁性膽管炎(primary biliary cholangitis,PBC)的藥物。過去的研究認(rèn)為UDCA可以通過FXR介導(dǎo)正向調(diào)控肝臟毛細(xì)膽管側(cè)膽汁酸轉(zhuǎn)運(yùn)體的表達(dá);然而有研究表明UDCA在人體內(nèi)起到抑制FXR的作用,這使得UDCA促進(jìn)肝膽汁酸分泌的作用機(jī)制仍有待明確。除了應(yīng)用于膽汁阻塞性疾病,UDCA及其衍生物在其他肝臟類疾病中也有所應(yīng)用,針對近年來發(fā)病率不...
