ERK1/2調(diào)控Drp1影響線粒體分裂參與癲癇發(fā)病機(jī)制的研究

目的:通過(guò)ERK1/2抑制劑(PD98059)對(duì)實(shí)驗(yàn)大鼠癲癇發(fā)作及Drp1表達(dá)變化的影響,探討ERK1/2通過(guò)調(diào)控線粒體分裂參與癲癇發(fā)作的可能機(jī)制。方法:將健康成年雄性SD大鼠分為以下幾組:正常組;癲癇組(0.5h、2h、6h、12h、24h六個(gè)時(shí)間點(diǎn));PD98059組(ERK1/2抑制劑);Mdivi-1抑制劑組(Drp1抑制劑組);二甲亞砜對(duì)照組(DMSO組)(每組或時(shí)間點(diǎn)n=5);通過(guò)腹腔注射氯化鋰-匹羅卡品建立癲癇模型,造模成功后,行為學(xué)評(píng)估大鼠首次發(fā)作的潛伏期及1小時(shí)之內(nèi)大鼠癲癇大發(fā)作的次數(shù)(Racine評(píng)分在IV級(jí)及以上則視為癲癇大發(fā)作),通過(guò)Western blot技術(shù)檢測(cè)分析ERK1/2和Drp1總蛋白及磷酸化蛋白在大鼠海馬中的表達(dá)水平,免疫組化觀察ERK1/2和Drp1在神經(jīng)細(xì)胞中的定位。結(jié)果:1.行為學(xué)結(jié)果:正常組大鼠無(wú)癲癇發(fā)作,癲癇組首次發(fā)作的潛伏期為15.4±1.14 min,PD98059組為32.8±5.54min;Mdivi-1組為28.6±1.94min;溶劑對(duì)照組為16.0±1.58min;癲癇組1小時(shí)之內(nèi)大鼠癲癇大發(fā)作的次數(shù)為9.8±0.83min...
