精子DNA完整性與基因甲基化及時(shí)間節(jié)律的相關(guān)研究

研究背景:精子DNA完整性的損傷是現(xiàn)代男性常見(jiàn)的精液異常,是導(dǎo)致男性生育力下降的主要因素。精子DNA碎片指數(shù)(DNA fragmentation index,DFI)是判斷精子DNA損傷的常用指標(biāo)。目前對(duì)于特定基因甲基化以及時(shí)間節(jié)律變化與精子DNA完整性的關(guān)系仍不清楚。第一部分 人精子印跡基因的異常甲基化與DNA損傷的關(guān)系研究目的:分析精子DNA損傷與印跡基因甲基化的相關(guān)性。方法:收集66例人精液樣本,精子染色質(zhì)結(jié)構(gòu)分析測(cè)定DFI,并檢測(cè)印跡基因H19、IGF2、KCNQ1、MEG3、MEST、PEG3和SNRPN 的甲基化水平,分析其甲基化與精子DNA損傷的關(guān)系。結(jié)果:與低DFI組相比,高DFI組精子印跡基因IGF2(cg17037101)位點(diǎn)的甲基化顯著降低(13.7±3%vs.31.5±5.3%,P=0.0053)。而其余印跡基因的甲基化水平無(wú)顯著變化。相關(guān)性分析表明,IGF2(cg17037101)甲基化水平與精子 DFI 呈負(fù)相關(guān)(r=-0.448,P=0.0038),而KCNQ1(cg24932449)甲基化水平與精子DFI呈正相關(guān)(r=0.354,P=0.0273)。結(jié)論...
