TLR4基因多態(tài)性與食管癌遺傳易感性關(guān)系的研究

目的Toll樣受體4(TLR4)是先天性免疫和獲得性免疫中的關(guān)鍵調(diào)控因子,與多種癌癥發(fā)生發(fā)展相關(guān),但關(guān)于TLR4基因多態(tài)性與食管癌易感性之間的關(guān)系少見報道。本研究旨在探討TLR4基因單核苷酸多態(tài)性(Single nucleotide polymorphism,SNP)對中國人群食管癌發(fā)病風(fēng)險的影響,篩選食管癌潛在的易感性生物標(biāo)志物。方法研究采用病例對照的方法,收集了480例食管癌患者標(biāo)本作為病例組,480例同期健康體檢者標(biāo)本作為對照組。經(jīng)過對數(shù)據(jù)庫進行分析,篩選出TLR4(rs1927914,rs11536891,rs7873784)3個SNPs作為研究靶點。分別采用聚合酶鏈?zhǔn)椒磻?yīng)-限制性片段長度多態(tài)性(Polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)技術(shù)和Taqman探針法進行基因分型。用χ2檢驗比較性別、年齡和吸煙狀況等相關(guān)因素的分布差異;用非條件Logistic回歸模型計算OR值及95%可信區(qū)間,以評價3個SNPs基因突變與食管癌發(fā)病風(fēng)險的關(guān)系。結(jié)果對比T...

圖 3 TLR4 rs7873784 基因多態(tài)性位點等位基因分型結(jié)果Fig.3 Genotyping result of TLR4 rs7873784 polymorphism1.2.2 研究對象一般人口學(xué)資料該病例對照研究包括食管癌患者和健康對照者各 480 例,具體見表 2。病例組與對照組中年齡、性別頻數(shù)分布均無統(tǒng)計學(xué)差異(P>0.05);病例組中吸煙者所占比例為 64.4%,明顯高于對照組,P 值小于 0.05,具有統(tǒng)計學(xué)差異。然而累計吸煙量在病例組與對照組中的分布無統(tǒng)計學(xué)差異(P=0.149)。1.2.3 TLR4 基因遺傳變異與食管癌易感性的關(guān)系TLR4 rs1927914、rs11536891 和 rs7873784 的基因型分布與食管癌的易感性關(guān)系見表 3。對照組中的 3 個 SNP 各基因型分布均符合 Hardy-Weinberg 遺傳平衡定律,說明所選擇人群具有良好的代表性。病例組中攜帶 TLR4 rs1927914 AA 基因型的個體為 195 例(40.6%),攜帶 GA 基因型的個體為 237 例(49.4%),攜帶
