結(jié)腸腺癌的發(fā)病機(jī)制探索及趨化因子CXC亞家族基因在結(jié)腸腺癌中的臨床應(yīng)用價(jià)值及機(jī)制研究

本研究分為四個(gè)部分:1、基于TCGA數(shù)據(jù)庫(kù)的結(jié)腸腺癌差異基因及預(yù)后相關(guān)基因生物信息學(xué)分析;2、基于TCGA數(shù)據(jù)庫(kù)的結(jié)腸癌患者趨化因子CXC亞家族基因表達(dá)的診斷和預(yù)后價(jià)值分析;3、廣西醫(yī)科大學(xué)一附院隊(duì)列中CXCL8、CXCL3和CXCL1表達(dá)在結(jié)腸腺癌中的診斷與預(yù)后價(jià)值分析;4、利用小RNA干擾技術(shù),分析SW620、RKO結(jié)腸癌細(xì)胞株中的CXCL8基因的生物學(xué)功能。第一部分 基于TCGA數(shù)據(jù)庫(kù)的結(jié)腸腺癌差異基因及預(yù)后相關(guān)基因生物信息學(xué)分析目的:通過(guò)分析癌癥基因組圖譜(TCGA)數(shù)據(jù)庫(kù)中結(jié)腸腺癌(COAD)的全基因組表達(dá)芯片數(shù)據(jù)集,鑒定COAD發(fā)生發(fā)展過(guò)程中的基因特征并評(píng)估其作為腫瘤生物標(biāo)志物的潛力。材料與方法:從TCGA數(shù)據(jù)庫(kù)下載COAD相關(guān)的mRNA測(cè)序數(shù)據(jù)集。使用R平臺(tái)的edgeR和DESeq軟件包篩選出COAD癌組織和癌旁正常結(jié)腸組織差異表達(dá)基因并用韋恩圖過(guò)濾出交集,再用R平臺(tái)的surviva1軟件包校正影響因子后計(jì)算出與COAD預(yù)后相關(guān)的基因,用韋恩圖過(guò)濾出與預(yù)后有關(guān)的差異基因,然后將這些不同作用的基因集合進(jìn)行基因功能富集分析、信號(hào)通路富集分析、構(gòu)建蛋白間和基因間相互作用網(wǎng)絡(luò)等生...
