肝細胞癌差異基因分析及長鏈非編碼RNA的初步驗證 ——基于轉(zhuǎn)錄組芯片的研究

研究目的和意義肝細胞癌(Hepatocellular carcinoma,HCC)是世界上高發(fā)的惡性腫瘤之一,其死亡率居惡性腫瘤的第三位。在中國,肝細胞癌的新發(fā)病例數(shù)和死亡率高居世界前列,其中,乙型肝炎病毒感染占大多數(shù)。在廣西地區(qū),肝細胞癌是死亡率最高的惡性腫瘤。由于肝細胞癌起病隱匿、腫瘤生長迅速、轉(zhuǎn)移性高,多數(shù)患者就診時已經(jīng)屬于中晚期。雖然目前肝臟手術(shù)切除或肝移植等治療手段可緩解病人情況,但是術(shù)后腫瘤復發(fā)和轉(zhuǎn)移嚴重影響患者的生活質(zhì)量。因此,深入研究肝細胞癌的發(fā)生、發(fā)展機制,探索針對肝細胞癌早期、特異性診斷標志物和治療靶點十分必要,這將為肝細胞癌的診斷和治療提供有力依據(jù)。肝細胞癌是一種高度異質(zhì)性的基因源性腫瘤。腫瘤基因本身不僅可導致細胞癌變,而且可通過表觀遺傳學水平調(diào)控引起肝細胞的癌變,主要包括甲基化、乙;、基因印記、染色體修飾和非編碼RNA調(diào)控等。其中,最近的研究證實,長鏈非編碼RNA(long noncoding rnas,lnc RNAs)在惡性腫瘤發(fā)生、發(fā)展中起到重要作用,如lnc RNA PCA,HOTAIR,H19等。長鏈非編碼RNA是一類長度大于200nt核苷酸RNA,...
