豬瘟病毒基因2型流行毒株和疫苗C株抗血清與相應(yīng)E2蛋白交叉免疫反應(yīng)性分析

【摘要】:為研究豬瘟病毒(CSFV)2.1基因亞型病毒株和疫苗C株E2蛋白與相應(yīng)抗血清的免疫反應(yīng)性差異,本研究在分析4株CSFV浙江流行株(QZ-14、JH-14、QZ-07和HZ-08)與疫苗C株E2變異程度的基礎(chǔ)上,制備了流行病毒株和疫苗C株全病毒豬抗血清以及針對(duì)流行株E2的單克隆抗體(MAb)4F4,比較了它們與同源株和異源株重組桿狀病毒表達(dá)的E2蛋白的親和性。結(jié)果顯示,4株浙江分離株中2株(QZ-14和JH-14)屬于亞型2.1d,另2株(QZ-07和HZ-08)為亞型2.1b,它們與C株E2的核苷酸同源性在82%~84%;間接免疫熒光鑒定顯示MAb 4F4能夠識(shí)別4株流行株,但不與C株反應(yīng);接種流行株的動(dòng)物均出現(xiàn)不同程度臨床癥狀;ELISA分析顯示疫苗C株血清與流行株E2蛋白的反應(yīng)性約為C株E2蛋白的27%~49%,2株流行株感染血清與C株E2蛋白反應(yīng)性約為流行株E2蛋白的34%~50%,免疫印跡分析顯示類似結(jié)果。以上結(jié)果表明CSFV基因2型流行株與疫苗C株E2蛋白的差異顯著影響其與相應(yīng)抗血清的交叉免疫反應(yīng)性,這一現(xiàn)象是否與C株豬瘟疫苗免疫保護(hù)效果下降有關(guān)還有待于進(jìn)一步研究。
[Abstract]:In order to study the difference of E2 protein between swine fever virus strain (CSFV) 2.1 and vaccine C strain E2 protein and corresponding antiserum, four strains of CSFV Zhejiang epidemic strain (QZ-14,JH-14,QZ-07 and HZ-08) and vaccine C strain E2 were prepared on the basis of analyzing the variation degree of E2 between epidemic virus strain and vaccine C strain. The swine antiserum of epidemic virus strain and vaccine C strain and the monoclonal antibody (MAb) 4F4 against epidemic strain E2 were prepared. The affinity between them and the E2 protein expressed by recombinant baculovirus was compared. The results showed that 2 of the 4 isolates in Zhejiang (QZ-14 and JH-14) belonged to subtypes 2.1 days, and the other two strains (QZ-07 and HZ-08) were subtypes 2.1b, and their nucleotides homology with E2 of strain C was 82% 鈮,
