健脾解毒方對大腸癌腫瘤血管形成相關基因表達的影響
[Abstract]:Objective: to investigate the effect of Jianpi jiedu recipe on the expression of angiogenic genes in colorectal cancer. Methods: the crude extract of Jianpi jiedu recipe was prepared by water extraction. The effect of Jianpi jiedu recipe crude extract on the proliferation of colorectal cancer HT29 cells was detected by MTT method. The IC50 (50%inhibitory concentration) value was calculated by, Graph Pad Prism 5 software. Affymetrix gene expression profile microarray was used to detect the differentially expressed genes after the intervention of Jianpi jiedu recipe and the path enrichment analysis was carried out. Based on the mammal rapamicin target protein (mammalian target of rapamycin, MTOR) the signal pathway uses IPA (Ingenuity Pathway Analysis) software to analyze the differentially expressed genes related to tumor angiogenesis and construct the network diagram. Western staining was used to verify the protein expression of key genes related to tumor angiogenesis. Results: the crude extract of Jianpi jiedu recipe could inhibit the proliferation of colorectal cancer HT29 cells. The IC50 values of 24 h, 48 h and 72 h after treatment were 13.060. 9.646 and 8.448 mg/m L.Affymetrix gene expression profiles were used to detect 218 genes significantly upregulated and 252 genes down-regulated after Jianpi Jiedu recipe intervention, mainly biosynthesis. Metabolism, apoptosis, antigen extraction, angiogenesis and other related genes, Among them, 12 differentially expressed genes related to tumor angiogenesis were analyzed by IPA software. The results of IPA software analysis showed that the intervention of Jianpi jiedu recipe could make sphingomyosphatide phosphodiesterase 3 (sphingomyelin phosphodiesterase 3, SMPD 3), VEGF, in colorectal cancer. Vascular endothelial growth factor A (vascular endothelial growth factor A, VEGFA), integration number 偽 subunit 1 (integrin subunit alpha 1, cathepsin B (cathepsin B, CTSB), cathepsin S (cathepsin S, CTSS) were down-regulated. GRB2 related binding protein 2 (GRB2 associated binding protein 2, GABA 2), PLAUR (plasminogen activator,urokinase receptor) and other genes were up-regulated. Western blot analysis showed that Jianpi jiedu recipe could significantly down-regulate the phosphorylation of mTOR (phospho-mTOR,P-mTOR). The expression of signal transduction and transcription activating factor 3 (signal transducer and activator of transcription 3), hypoxia inducible factor 1 偽 (hypoxia inducible factor-1 偽 (HIF-1 偽) and VEGF protein were significantly up-regulated by phosphorylation of p53 (phospho-P53,P-P53) protein. Conclusion: Jianpi jiedu recipe may inhibit tumor angiogenesis of colorectal cancer by affecting mTOR-HIF-1 偽-VEGF signal pathway, which may be one of the mechanisms of Jianpi jiedu recipe in the treatment of colorectal cancer.
