近交對(duì)馬氏珠母貝生長(zhǎng)性狀、遺傳多樣性及礦化基因表達(dá)的影響

【摘要】:【目的】探討近交對(duì)馬氏珠母貝生長(zhǎng)性狀、遺傳結(jié)構(gòu)及礦化基因表達(dá)的影響,闡明近交引起子一代性狀退化的遺傳原理,為貝類品種培育提供參考依據(jù)!痉椒ā恳詢蓚(gè)黃殼色全同胞家系(M和N)為親本,按照因子設(shè)計(jì)構(gòu)建4個(gè)家系組合(F1:M1♀×M1♂;F2:M1♀×N1♂;F3:N1♀×M1♂;F4:N1♀×N1♂),比較4個(gè)家系成貝生長(zhǎng)性狀差異,利用9對(duì)SSR引物分析其遺傳多樣性,并以實(shí)時(shí)熒光定量PCR檢測(cè)家系間礦化基因nacrein、pearlin和pif177的表達(dá)差異!窘Y(jié)果】4個(gè)家系間的平均殼長(zhǎng)、平均殼高、平均殼寬、平均體重和平均殼重存在明顯差異,且近交家系(F1和F4)的平均殼長(zhǎng)、平均殼高、平均殼寬、平均體重和平均殼重均小于雜交家系(F2和F3),其生長(zhǎng)性狀退化率(ID)為11.7%~29.4%。家系F1、F2、F3和F4的平均觀測(cè)雜合度分別為0.2525、0.3116、0.3178和0.2752,平均期望雜合度分別為0.3451、0.3822、0.4005和0.3549,平均多態(tài)信息含量(PIC)分別為0.4591、0.5389、0.4878和0.4810。雜交家系(F2和F3)礦化基因nacrein、pearlin和pif177的相對(duì)表達(dá)量均高于近交家系(F1和F4),且4個(gè)家系的生長(zhǎng)性狀平均值與3個(gè)礦化基因的相對(duì)表達(dá)量存在明顯正相關(guān)!窘Y(jié)論】近交能導(dǎo)致馬氏珠母貝生長(zhǎng)性狀、遺傳多樣性及礦化基因表達(dá)量明顯降低,因此制定馬氏珠母貝育種方案時(shí)應(yīng)構(gòu)建合理的交配組合系統(tǒng),防止因近交導(dǎo)致性狀退化與遺傳多樣性指標(biāo)降低而影響育種進(jìn)程。
