利用改良SOE-PCR技術(shù)定點(diǎn)突變TuMVC4-VPg基因

【摘要】:蕪菁花葉病毒(Turnip mosaic virus,Tu MV)是白菜類蔬菜生產(chǎn)上的重要病害,其中Tu MV VPg基因在Tu MV侵染白菜類蔬菜的過程中起著至關(guān)重要的作用,且不同Tu MV株系的致病性不同。本試驗(yàn)采用改良的重疊延伸PCR(SOE-PCR)技術(shù),以Tu MV C4-VPg基因序列為模板,定點(diǎn)突變Tu MV C4-VPg與Tu MV CDN1-VPg基因間5個(gè)差異核苷酸位點(diǎn)(決定5個(gè)氨基酸差異)。結(jié)果表明:通過SOE-PCR技術(shù)獲得了5個(gè)Tu MV C4-VPg基因的單點(diǎn)突變體,即Tu MV C4-VPg-Ⅰ、Tu MV C4-VPg-Ⅱ、Tu MV C4-VPg-Ⅲ、Tu MV C4-VPg-Ⅳ和Tu MV C4-VPg-Ⅴ。
[Abstract]:Turnip mosaic virus (Turnip mosaic virus,Tu MV) is an important disease in cabbage vegetable production, in which Tu MV VPg gene plays an important role in the process of Tu MV infecting cabbage vegetables, and the pathogenicity of different Tu MV strains is different. In this experiment, an improved overlapping extension PCR (SOE-PCR) technique was used to determine the difference of 5 amino acids between site-directed mutant Tu MV C4-VPg and Tu MV CDN1-VPg gene using Tu MV C4-VPg gene sequence as template. The results showed that five single point mutants of Tu MV C4-VPg gene were obtained by SOE-PCR, that is, Tu MV C4 VPgI, Tu MV C4 VPgII, Tu MV C4 VPgIII, Tu MV C4 VPgIV and Tu MV C4 VPgV.
