【摘要】:MYB類轉(zhuǎn)錄因子在植物觀賞器官色彩形成中發(fā)揮關(guān)鍵作用。為了研究紅掌中該類轉(zhuǎn)錄因子的種類、表達(dá)、作用機理等,本研究從紅掌中獲得了一個MYB轉(zhuǎn)錄因子的基因序列,通過反轉(zhuǎn)錄多聚酶鏈?zhǔn)椒磻?yīng)(RT-PCR)、生物信息學(xué)軟件分析、熒光定量PCR檢測、構(gòu)建超表達(dá)載體并異源轉(zhuǎn)化煙草等手段,對該轉(zhuǎn)錄因子進(jìn)行了初步研究。結(jié)果表明,該轉(zhuǎn)錄因子編碼基因包含完整編碼區(qū),共計912 bp,編碼303個氨基酸殘基,序列組成與其他物種同源體具有高度相似性;熒光定量分析顯示,Aa MYB1在紅掌不同組織部位都有表達(dá),但在苞片中表達(dá)量最高;獲得了12株陽性轉(zhuǎn)化株,形態(tài)觀察發(fā)現(xiàn)轉(zhuǎn)化株營養(yǎng)器官花色素累積程度隨基因表達(dá)不同而異,但可使所有轉(zhuǎn)化株花器官顏色顯著加深。本研究結(jié)果為進(jìn)一步探究MYB轉(zhuǎn)錄因子在紅掌中調(diào)控花色素合成等信息提供了有益參考。
[Abstract]:MYB transcription factors play a key role in color formation of plant ornamental organs. In order to study the species, expression and mechanism of this kind of transcription factors in red palm, a gene sequence of MYB transcription factor was obtained from red palm, which was analyzed by reverse transcriptional polymerase chain reaction (RT-PCR) and bioinformatics software. Fluorescence quantitative PCR (FQ-PCR) was used to detect the transcription factor, and the overexpression vector was constructed and the tobacco was transformed from different sources, and the transcription factor was studied. The results showed that the transcriptional factor encoded gene contained a complete coding region, and a total of 912 bp, encoded 303amino acid residues, and the sequence composition was highly similar to that of other species. Fluorescence quantitative analysis showed that Aa MYB1 was expressed in different tissues of red palm, but the highest expression was found in bracts. Twelve positive transformed strains were obtained. morphological observation showed that the degree of anthocyanin accumulation in vegetative organs of transformed plants varied with gene expression, but the color of floral organs of all transformed plants was significantly deepened. The results of this study provide a useful reference for further exploring the information of MYB transcription factors regulating anthocyanin synthesis in red palm.
