水稻開花時間基因OsDTH10的過表達分析

【摘要】:開花時間(抽穗期)是農(nóng)作物一個重要的農(nóng)藝性狀,與農(nóng)作物的產(chǎn)量息息相關(guān)。調(diào)節(jié)農(nóng)作物的開花抽穗時間是提高農(nóng)作物產(chǎn)量的一條重要途徑。為了揭示水稻開花時間基因OsDTH10在水稻抽穗期調(diào)控中的功能,利用反向遺傳學(xué)方法構(gòu)建了水稻OsDTH10基因過量表達載體,獲得轉(zhuǎn)基因植株,分析目的基因在過量表達條件下的功能。結(jié)果表明,過量表達OsDTH10導(dǎo)致水稻的抽穗期推遲;RT-PCR檢測結(jié)果表明OsDTH10的表達量在晚抽穗的轉(zhuǎn)基因株系中明顯提高,但在沒有表型的轉(zhuǎn)基因株系及野生型中表達量較低,說明晚抽穗的表型是由于OsDTH10過量表達所致。組織特異性表達結(jié)果表明OsDTH10在植物的不同器官中都有表達,但在水稻的莖、葉鞘中表達量較高。分析OsDTH10在不同葉齡葉片中的表達,結(jié)果表明OsDTH10在未完全展開的劍葉葉片及倒2葉中的表達量較高,但在下部較老的倒3葉及倒4葉中的表達量下降。另外,分析OsDTH10在不同光周期條件下的晝夜節(jié)律性表達,結(jié)果表明無論在短日照還是長日照條件下,OsDTH10都在白天(光期)有表達高峰,在夜晚(暗期)表達降低,表明OsDTH10可能涉及光周期調(diào)控通路調(diào)節(jié)水稻的抽穗期。
[Abstract]:Flowering time (heading stage) is an important agronomic character of crops, which is closely related to crop yield. Adjusting the flowering and heading time of crops is an important way to increase crop yield. In order to reveal the function of rice flowering time gene OsDTH10 in the regulation of heading stage of rice, the over-expression vector of rice OsDTH10 gene was constructed by reverse genetics method, and the transgenic plants were obtained. The function of the target gene under the condition of over-expression was analyzed. The results showed that the overexpression of OsDTH10 resulted in delayed heading date of rice. The results of RT-PCR analysis showed that the expression of OsDTH10 was significantly increased in transgenic lines with late heading, but was lower in transgenic lines without phenotype and wild type, indicating that the phenotype of late heading was caused by over-expression of OsDTH10. The results of tissue-specific expression showed that OsDTH10 was expressed in different organs of plants, but it was higher in stem and leaf sheath of rice. The expression of OsDTH10 in the leaves of different leaf ages was analyzed. The results showed that the expression of OsDTH10 was higher in the leaves of sword leaves and 2 leaves of the other leaves, but decreased in the lower leaves of the older leaves of the third leaf and the other leaves of the other leaves. In addition, the circadian rhythmic expression of OsDTH10 under different photoperiod conditions was analyzed. The results showed that the expression of OsDTH10 reached a peak during daytime (light period) and decreased at night (dark phase), both under short and long sunshine conditions. These results suggest that OsDTH10 may be involved in the regulation of heading stage by photoperiod regulation pathway.
