紫蘇脂肪酸硫酯酶基因PfFatA生物信息學(xué)及其表達(dá)特性分析

【摘要】:為研究脂肪酸硫酯酶A(FatA)在紫蘇脂肪酸合成機(jī)制中發(fā)揮的作用,對紫蘇FatA基因及其編碼的蛋白質(zhì)進(jìn)行生物信息學(xué)分析。結(jié)果表明,PfFatA基因cDNA全長1 652 bp,開放閱讀框1 119 bp,編碼372個(gè)氨基酸;紫蘇FatA基因編碼的蛋白質(zhì)為親水性不穩(wěn)定蛋白,無跨膜區(qū)域;含Acyl-ACP_TE保守結(jié)構(gòu)域,屬于Hotdog fold超蛋白家族;多序列比對結(jié)果發(fā)現(xiàn),序列C端含有保守的三聯(lián)肽AKL和SKV結(jié)構(gòu);系統(tǒng)進(jìn)化樹分析表明,紫蘇與丹參親緣關(guān)系較近。通過Real-time PCR分析得知,PfFatA在紫蘇根、莖、葉、花和種子不同發(fā)育時(shí)期均有表達(dá),其中,在種子發(fā)育初期表達(dá)量最高。該研究結(jié)果可為進(jìn)一步闡明PfFatA基因的功能及作用機(jī)制奠定基礎(chǔ)。
[Abstract]:In order to study the role of fatty acid thioesterase A (FatA) in the fatty acid synthesis mechanism of Perilla perilla, bioinformatics analysis of perilla FatA gene and its encoded protein was carried out. The results showed that the full length cDNA of PfFatA gene was 1 652 bp, open reading frame (ORF) 1 119 bp, encoding 372 amino acids, the protein encoded by perilla FatA gene was a hydrophilic unstable protein with no transmembrane region. The results of multi-sequence alignment showed that the C-terminal of the sequence contained conserved tripeptide AKL and SKV, and phylogenetic tree analysis showed that the relationship between Radix Perilla and Salvia miltiorrhiza was close to that of Radix Perilla and Salvia miltiorrhiza (Salvia miltiorrhiza). Real-time PCR analysis showed that PfFatA was expressed in the root, stem, leaf, flower and seed of Perilla perilla at different developmental stages, and the highest expression was found in the early stage of seed development. The results of this study may lay a foundation for further elucidating the function and mechanism of PfFatA gene.
