【摘要】：以紅掌栽培品種‘Dakota’為材料,采用RACE技術克隆獲得An CHI基因的c DNA序列,分析花青素含量與該基因表達的關系,進一步完善紅掌花青素合成途徑。結(jié)果表明:An CHI序列全長為1 117 bp,其中開放閱讀框為777 bp,編碼258個氨基酸;An CHI在不同時期紅掌佛焰苞以及根、莖、葉、肉穗組織中均有表達,且在佛焰苞發(fā)育前期表達量明顯高于后期,測得花青素含量與基因表達量變化基本一致;構建的p GEX-4T1-An CHI重組表達載體在原核中正確表達。初步驗證了An CHI的表達可能對紅掌佛焰苞花青素的合成有一定促進作用。
[Abstract]:The c-DNA sequence of An CHI gene was cloned by RACE using 'Dakota' as material. The relationship between anthocyanin content and the expression of An CHI gene was analyzed, and the synthesis pathway of anthocyanidin was further improved. The results showed that the full length of: An CHI was 1 117 bp, and the open reading frame (ORF) was 777 bp, encoding 258 amino acids. An CHI was expressed in the flame bud, root, stem, leaf and panicle of red palm at different stages, and the expression level was significantly higher in the early stage of development than in the later stage, and the content of anthocyanin was basically the same as the gene expression in the early stage of the development of the flame bud. The recombinant expression vector of p GEX-4T1-An CHI was expressed correctly in E. coli. The expression of An CHI may play an important role in the synthesis of anthocyanidins.
