3個(gè)白樺BpBEE基因的克隆與表達(dá)分析

【摘要】:根據(jù)已有的白樺Betula platyphylla莖尖、葉片及木質(zhì)部等45個(gè)轉(zhuǎn)錄組信息,獲得3條白樺BEE基因的全長(zhǎng)c DNA序列,分別命名為BpBEE1,BpBEE2和BpBEE3。對(duì)3個(gè)BpBEE基因進(jìn)行了生物信息學(xué)分析和實(shí)時(shí)定量聚合酶鏈?zhǔn)椒磻?yīng)分析,結(jié)果表明:3個(gè)基因具有典型的結(jié)合DNA及特異識(shí)別序列E-box和N-box,屬于b HLH類轉(zhuǎn)錄因子。這3個(gè)基因與擬南芥Arabidopsis thaliana中At BEE基因親緣關(guān)系較近,屬于b HLH超家族第25亞類。在白樺生長(zhǎng)季內(nèi),BpBEE1,BpBEE2和BpBEE3在不同組織中均有表達(dá),葉中BpBEE基因表達(dá)量明顯高于其他部位。在生長(zhǎng)旺盛時(shí)期,莖、葉和頂芽中BpBEE基因均呈現(xiàn)上調(diào)表達(dá),表明BpBEE基因可能參與到白樺的生長(zhǎng)過程。油菜素內(nèi)酯(BR)激素處理之后,BpBEE1在早期(2 h和4 h)的芽和木質(zhì)部中呈顯著上調(diào)(2倍);BpBEE2在早期(2 h)的木質(zhì)部和芽顯著上調(diào)外,其他處理時(shí)間也呈現(xiàn)不同程度的上調(diào)。表明3個(gè)白樺BpBEE基因參與BR激素應(yīng)答,且BpBEE1和BpBEE2呈現(xiàn)早期應(yīng)答響應(yīng)。
[Abstract]:According to the information of 45 transcription groups such as stem tip, leaf and xylem of Bai Hua Betula platyphylla, the full length c DNA sequence of Bai Hua BEE gene was obtained, which was named BpBEE1,BpBEE2 and BpBEE3. respectively. Three BpBEE genes were analyzed by bioinformatics and real-time quantitative polymerase chain reaction (QPCR). The results showed that the three genes had typical binding DNA and specific recognition sequences E-box and Nkbox, and belonged to b-HLH transcription factor. These three genes are closely related to At BEE gene in Arabidopsis thaliana, and belong to b-HLH superfamily subgroup 25. During the growing season of Bai Hua, both BpBEE1,BpBEE2 and BpBEE3 were expressed in different tissues, and the expression of BpBEE gene in leaves was significantly higher than that in other parts. During the period of vigorous growth, the expression of BpBEE gene was up-regulated in stem, leaf and terminal bud, suggesting that BpBEE gene might be involved in the growth process of Bai Hua. After treatment with (BR) hormone, BpBEE1 was up-regulated significantly in bud and xylem of early stage (2 h and 4 h) (2 times). BpBEE2 was significantly up-regulated in xylem and bud at early stage (2 h), and other treatment time was also up-regulated in varying degrees. The results showed that three Bai Hua BpBEE genes were involved in BR hormone response, and BpBEE1 and BpBEE2 showed early response.
