CPPU對‘陽光玫瑰’葡萄品質及香氣合成相關基因表達的影響

【摘要】:[目的]研究不同質量濃度細胞分類素(CPPU)對葡萄果實香氣組分及合成相關基因表達的影響,為闡明香氣物質的合成提供理論依據(jù)。[方法]以‘陽光玫瑰’葡萄為材料,在花期時用25 mg·L~(-1)GA_3處理花序,花后2周時用25 mg·L~(-1)GA_3+0 mg·L~(-1)CPPU處理果穗作為對照(CK),設置25 mg·L~(-1)GA_3+5 mg·L~(-1)CPPU(Ⅰ)、25 mg·L~(-1)GA_3+10 mg·L~(-1)CPPU(Ⅱ)、25 mg·L~(-1)GA_3+15 mg·L~(-1)CPPU(Ⅲ)3個處理,采用實時熒光定量PCR法研究香氣合成相關基因脫氧木酮糖磷酸合酶基因(DXS3)、香葉基二磷酸合酶基因(GPPS)和里那醇合成酶基因(LIS)的表達變化,并利用GC-MS技術測定葡萄香氣組分。[結果]與對照相比,處理Ⅰ和Ⅱ的果實顯著增大、增重,可滴定酸含量和可溶性固形物有所下降,外觀品質提高,處理Ⅲ因CPPU濃度過高而引起葡萄品質下降。與對照相比,香氣種類減少,萜類物質特征香氣里那醇、順式氧化里那醇、香葉醇等相對含量降低,風味變淡,且CPPU濃度過高,不利于香氣的產(chǎn)生。與對照相比,經(jīng)不同濃度CPPU處理的果實中香氣合成相關基因DXS3、GPPS、LIS表達量高峰推遲且峰值下調,濃度越高,推遲時間越久,峰值更低,基因表達受抑制,這與萜類物質相對含量降低相一致。[結論]25 mg·L~(-1)GA_3+5 mg·L~(-1)CPPU處理能夠改善‘陽光玫瑰’葡萄外觀品質并保證內在品質。
