犬瘟熱病毒H基因的序列分析

【摘要】:【目的】探索犬瘟熱病毒(Canine distemper virus,CDV)基因的遺傳變異情況,為CDV的防控提供理論依據(jù)�!痉椒ā渴占�2014-2015年流行于上海和安徽兩地的CDV野毒株,用RT-PCR方法克隆其血球凝集素蛋白基因(H),從分子水平上討論CDV H基因的流行規(guī)律、遺傳進化特性和變異情況。【結(jié)果】分離的5株CDV之間H基因核苷酸和氨基酸序列的同源性分別為97.5%~99.9%和97.5%~99.2%,其與CDV疫苗株CDV3和Onderstepoort H基因核苷酸的同源性為90.9%~99.9%,氨基酸的同源性為90.4%~99.6%。進化樹分析結(jié)果表明,分離到的5株CDV野毒株均屬于亞洲Ⅰ型,與大部分的亞洲Ⅰ型處于同一分支。CDV H基因有9處潛在的天冬氨酸糖基化位點;H基因整體的同義突變概率與非同義突變概率的比例,即ds/dn=5.887 0�!窘Y(jié)論】選擇壓力并未作用于分離到的CDV毒株,而是在中性選擇作用下進化的。

[methods] to collect the field strains of canine distemper virus (CDV) prevalent in Shanghai and Anhui from 2014 to 2015. The RT-PCR method was used to clone the hemagglutinin protein gene (H),. The prevalence of CDV H gene was discussed at the molecular level. Genetic evolution and variation. [results] the homology of nucleotide and amino acid sequences of H gene between the five CDV isolates were 97.5% and 99.2%, respectively. The homology of nucleotide sequence with CDV3 and Onderstepoort H gene of CDV vaccine strain was 99. 9, and the homology of amino acid with CDV3 and Onderstepoort H gene was 94. 4 and 99. 6 respectively.
