2015年黑龍江省豬流行性腹瀉病毒ORF3和S1基因的遺傳變異分析

【摘要】:為了解黑龍江省豬流行性腹瀉病毒(PEDV)流行現(xiàn)狀及遺傳進(jìn)化情況,試驗(yàn)利用RT-PCR方法對2015年在黑龍江省采集的35份PEDV陽性病料進(jìn)行檢測,針對PEDV ORF3和S1基因進(jìn)行擴(kuò)增及測序,并對其進(jìn)行遺傳進(jìn)化分析。結(jié)果表明:ORF3基因和S1基因的核苷酸同源性分別為98.8%~100%和96.6%~100%,其中HLJ2015/DP1-1毒株與經(jīng)典毒株CV777相比,S1基因在1 130~1 141 bp之間有12個(gè)核苷酸缺失;S1基因系統(tǒng)進(jìn)化樹分析顯示,PEDV S1基因分為GⅠ群和GⅡ群,此次試驗(yàn)鑒定的PEDV毒株屬于GⅡ群的non S-INDEL型毒株。
[Abstract]:In order to understand the epidemic status and genetic evolution of porcine epidemic diarrhea virus (PEDV) in Heilongjiang Province, 35 samples of PEDV positive venereal diseases collected in Heilongjiang Province in 2015 were detected by RT-PCR method. PEDV ORF3 and S1 genes were amplified and sequenced, and their genetic evolution was analyzed. The results showed that the nucleotide homology of ORF3 gene and S1 gene were 98.8% and 96.6%, respectively. Compared with classical strain CV777, S1 gene had 12 nucleotide deletions between HLJ2015/DP1-1 strain and classical strain CV777.
