【摘要】：目的:探討含反藥組合的海藻玉壺湯中海藻不同品種與甘草加減應(yīng)用對(duì)甲狀腺腫大大鼠肝臟CYP450酶基因與蛋白表達(dá)的影響。方法:將大鼠隨機(jī)分為空白組、模型組、陽(yáng)性藥組、海蒿子全方組、羊棲菜全方組、海蒿子去甘草組、羊棲菜去甘草組、去海藻組、去海藻甘草組,除空白組外其余各組灌服丙硫氧嘧啶復(fù)制甲狀腺腫大模型,以優(yōu)甲樂(lè)作為陽(yáng)性對(duì)照,各給藥組分別給予相應(yīng)藥物。檢測(cè)各組大鼠CYP2E1、CYP3A1、CYP3A2mRNA表達(dá)及CYP2E1蛋白表達(dá)。結(jié)果:與空白組比較,模型組CYP2E1/3A2 mRNA表達(dá)明顯升高(P0.05),CYP3A1 mRNA表達(dá)、CYP2E1蛋白表達(dá)有升高趨勢(shì);陽(yáng)性藥組CYP3A1 mRNA表達(dá)明顯升高(P0.05)。與模型組比較,陽(yáng)性藥組、海蒿子全方組、羊棲菜全方組CYP2E1 mRNA表達(dá)明顯降低(P0.05);羊棲菜全方組、去海藻組CYP3A2 mRNA表達(dá)明顯降低(P0.05)。結(jié)論:丙硫氧嘧啶所致甲狀腺腫大大鼠肝臟CYP2E1/3A1/3A2 mRNA表達(dá)增強(qiáng),可能會(huì)對(duì)肝臟內(nèi)的前毒物和前致癌物有一定的誘導(dǎo)作用,并且促進(jìn)藥物代謝吸收,從而引起肝損傷;在這一病理狀態(tài)下,海藻玉壺湯(海蒿子/羊棲菜)全方對(duì)增強(qiáng)的相關(guān)基因表達(dá)有回調(diào)作用,并且顯示出對(duì)相關(guān)蛋白表達(dá)的抑制作用,從而抑制肝臟內(nèi)的前毒物,抑制藥物吸收,發(fā)揮保護(hù)肝功能的作用。
[Abstract]:Aim: to investigate the effects of different seaweed varieties and liquorice on the expression of CYP450 gene and protein in the liver of rats with thyroid enlargement. Methods: rats were randomly divided into blank group, model group, positive drug group, whole prescription group, licorice group and seaweed group. The model of goiter was made by oral administration of prothiouracil in all groups except the blank group. The expression of CYP2E1,CYP3A1,CYP3A2mRNA and CYP2E1 protein were detected. Results: compared with the blank group, the expression of CYP2E1/3A2 mRNA was significantly higher in the model group (P0.05), the expression of CYP3A1 mRNA and CYP2E1 protein was higher than that of the control group (P0.05), and the expression of CYP3A1 mRNA in the positive drug group was significantly higher (P0.05). Compared with the model group, the expression of CYP2E1 mRNA was significantly decreased in the positive drug group, Artemisia artemisia group and whole prescription group (P0.05); the CYP3A2 mRNA expression in the whole prescription group and seaweed group was significantly decreased (P0.05). Conclusion: the increased expression of CYP2E1/3A1/3A2 mRNA in the liver of rats induced by propylthiouracil may induce pretoxic and precancerous substances in the liver, and promote the absorption of drug metabolism, which may lead to liver injury. In this pathological state, the whole prescription of seaweed jug (Artemisia annua / Sargassum chinensis) has a callback on the enhanced expression of related genes, and has shown an inhibitory effect on the expression of related proteins, thereby inhibiting the pretoxic substances in the liver. Inhibition of drug absorption, play a role in protecting liver function.
