水牛6個Y染色體基因的拷貝數(shù)變異研究

【摘要】:目前,牛Y染色體轉(zhuǎn)錄組學(xué)分析表明至少有6種編碼蛋白的多拷貝基因位于Y染色體雄性特異區(qū)(MSY)。牛MSY基因的拷貝數(shù)變異與雄性繁殖力和精子發(fā)生機制相關(guān)。由于目前沒有水牛全基因組序列,國內(nèi)外僅研究了水牛Y染色體上TSPY基因的拷貝數(shù)變異。關(guān)于Y染色體多拷貝基因拷貝數(shù)變異研究,主要集中在人類、牛及小鼠的TSPY、HSFY及PRAMEY等基因,沒有關(guān)于水牛6個Y染色體基因HSFY、ZNF280BY、SRY、DBY、UTY和OFD1Y拷貝數(shù)變異的報道。因此,本研究使用實時定量PCR(q PCR)方法,檢測3個Y染色體基因SRY、HSFY和ZNF280BY在16個中國沼澤型水牛品種、外國沼澤型水牛(越南水牛和老撾水牛)及江河型水牛(摩拉水牛和尼里拉菲水牛)共計302頭公水牛中的拷貝數(shù)變異,以及3個Y染色體基因DBY、UTY和OFD1Y在6個水牛品種105頭公水牛中的拷貝數(shù)變異,獲得如下創(chuàng)新性結(jié)果:1.首次揭示水牛6個Y染色體基因(HSFY、ZNF280BY、SRY、DBY、UTY和OFD1Y基因)的拷貝數(shù)變異,發(fā)現(xiàn)HSFY,ZNF280BY和DBY基因的拷貝數(shù)中值數(shù)(變化范圍)分別為48(20-145),257(73-974)和2(1-4);SRY和OFD1Y基因的拷貝數(shù)中值數(shù)(變化范圍)為1(1-4),在越南水牛中拷貝數(shù)表現(xiàn)出差異;UTY基因拷貝數(shù)為1。2.揭示HSFY和ZNF280BY基因在18個水牛品種中為多拷貝基因,且拷貝數(shù)在不同個體和品種中有很大差異;DBY基因在6個水牛品種中為低拷貝基因;SRY、OFD1Y和UTY基因在中國水牛品種中為單拷貝基因。3.發(fā)現(xiàn)HSFY、ZNF280BY、SRY、DBY、UTY和OFD1Y基因的拷貝數(shù)變異在沼澤型水牛和江河型水牛中沒有顯著差異。
[Abstract]:At present, transcriptome analysis of bovine Y chromosome indicates that at least 6 multicopy genes of protein are located in the male specific region of Y chromosome (MSY). The copy number variation of bovine MSY gene is related to male reproduction and spermatogenesis. Because there is no buffalo genome sequence at present, only the copy number variation of TSPY gene on buffalo Y chromosome has been studied at home and abroad. The study on the copy number variation of Y chromosome multiple copy genes is mainly focused on the TSPY,HSFY and PRAMEY genes of human, cattle and mice. There is no report on the HSFY,ZNF280BY,SRY,DBY,UTY and OFD1Y copy number variation of six Y chromosome genes in buffalo. Therefore, real-time quantitative PCR (q PCR) was used to detect three Y chromosome genes SRY,HSFY and ZNF280BY in 16 Chinese marsh buffalo breeds. Copy number variation and DBY, of three Y chromosome genes in 302 foreign swamp buffalo (Vietnamese buffalo and Lao buffalo) and river water buffalo (Mora buffalo and Nilirafi buffalo) The copy number variation of UTY and OFD1Y in 105 male buffalo breeds was as follows: 1. The variation of copy number of six Y chromosome genes (HSFY,ZNF280BY,SRY,DBY,UTY and OFD1Y) in buffalo was revealed for the first time. It was found that the median copy number (range of variation) of HSFY,ZNF280BY and DBY genes was 48 (20-145), respectively. 257 (73-974) and 2 (1-4); The median copy number (range of variation) of SRY and OFD1Y gene was 1 (1-4), which was different in Vietnamese buffalo, and the copy number of UTY gene was 1.2. The results showed that HSFY and ZNF280BY genes were multi-copy genes in 18 buffalo breeds, and the number of copies was different in different individuals and varieties, and DBY gene was low copy gene in 6 buffalo breeds. SRY,OFD1Y and UTY genes are single copy genes in Chinese buffalo breeds. It was found that there was no significant difference in copy number variation of HSFY,ZNF280BY,SRY,DBY,UTY and OFD1Y gene between swamp buffalo and river buffalo.
