轉(zhuǎn)錄因子7類似物-2基因rs12255372多態(tài)性與妊娠期糖尿病發(fā)病風(fēng)險(xiǎn)的系統(tǒng)評(píng)價(jià)

【摘要】:目的探討轉(zhuǎn)錄因子7類似物-2(TCF7L2)基因rs12255372 GT多態(tài)性與妊娠期糖尿病(GDM)發(fā)病風(fēng)險(xiǎn)的關(guān)系。方法截止2016年9月,檢索NCBI Pub Med、中國(guó)知網(wǎng)(CNKI)及萬(wàn)方等數(shù)據(jù)庫(kù)關(guān)于rs12255372多態(tài)性與GDM發(fā)病相關(guān)病例-對(duì)照研究。Stata12.0軟件檢測(cè)所納入數(shù)據(jù)發(fā)表偏倚及異質(zhì)性,以合并效應(yīng)的OR及95%置信區(qū)間(95%CI)評(píng)估rs12255372位點(diǎn)與GDM的發(fā)病風(fēng)險(xiǎn)。結(jié)果依據(jù)納入、排除標(biāo)準(zhǔn),共9篇文獻(xiàn),包含5778例研究對(duì)象納入本研究。Meta分析顯示,與G基因型相比,T等位基因攜帶者GDM發(fā)病風(fēng)險(xiǎn)顯著增高,整體合并效應(yīng)的OR值及95%CI為1.28(1.16~1.40);以種族進(jìn)行亞組分析,高加索人群合并效應(yīng)的OR及95%CI為1.29(1.15~1.45),亞洲人群為1.25(1.05~1.48)。基因型分析顯示,顯性遺傳模型下,rs12255372 GT與GDM發(fā)病顯著相關(guān),與GG基因型相比,GT+TT基因型攜帶者GDM發(fā)病風(fēng)險(xiǎn)顯著增高,整體合并效應(yīng)的OR及95%CI為1.35(1.20~1.53),高加索人群為1.39(1.20~1.62),亞洲人群為1.28(1.04~1.58)。結(jié)論 TCF7L2rs12255372 GT多態(tài)性與GDM發(fā)病密切相關(guān),G(T突變可能增加妊娠期婦女GDM的發(fā)病風(fēng)險(xiǎn)。
[Abstract]:Objective to investigate the relationship between the rs12255372 GT polymorphism of transcription factor 7 analogue 2 (TCF7L2) gene and the risk of gestational diabetes mellitus (GDM) (GDM). Methods as of September 2016, a case-control study on rs12255372 polymorphism and GDM was searched in NCBI Pub Med, China knowledge Network (CNKI) and Wanfang databases. The published bias and heterogeneity of the data included in the Stata12.0 software test were analyzed. OR and 95% confidence interval (95%CI) of combined effect were used to evaluate the risk of rs12255372 and GDM. Results according to inclusion and exclusion criteria, 9 articles including 5778 subjects were included in this study. Meta analysis showed that T allele carriers had significantly higher risk of GDM compared with G genotype. The OR and 95%CI of the whole merger effect were 1.28 (1.16 鹵1.40), and the OR and 95%CI of Caucasian population were 1.29 (1.151.45) and 1.25 (1.051.48) respectively. Genotypic analysis showed that rs12255372 GT was significantly correlated with GDM in dominant genetic model. Compared with GG genotype, the incidence of GDM in, GT TT genotype carriers was significantly higher than that in GG genotype. The overall combined effect of OR and 95%CI were 1.35 (1.201.53), 1.39 (1.201.62) in Caucasian population and 1.28 (1.041.58) in Asian population. Conclusion TCF7L2rs12255372 GT polymorphism is closely associated with GDM and, G (T mutation may increase the risk of GDM in pregnant women.
