甜蕎FaesAP2基因的克隆與表達分析

【摘要】:本研究采用同源克隆和RACE技術,從甜蕎(Fagopyrum esculentum Moench.)品種‘西農(nóng)9976’中分離出調(diào)控花器官發(fā)育的Faes AP2基因,該基因序列全長1668 bp,包含1個長為1374 bp的完整開放閱讀框,共編碼457個氨基酸。序列比對以及系統(tǒng)發(fā)育分析結(jié)果發(fā)現(xiàn),Faes AP2蛋白擁有2個高度保守的AP2(APETALA2)結(jié)構(gòu)域,在第1個AP2結(jié)構(gòu)域前端有1段由10個氨基酸殘基組成的高度保守的核定位信號區(qū);系統(tǒng)發(fā)育分析顯示其與擬南芥(Arabidopsis thaliana(L.)Heynh.)AP2轉(zhuǎn)錄因子的親緣關系較近;虮磉_模式分析表明,該基因在甜蕎pin型和thrum型花的雄蕊和雌蕊中有明顯的表達,但在幼葉和花被片中不表達,且其表達量在2種類型花不同發(fā)育時期呈現(xiàn)明顯的變化,均在花藥迅速膨大期達到最高值,因此推測該基因在甜蕎花發(fā)育過程中可能參與了花器官發(fā)育的調(diào)控。
[Abstract]:Faes AP2 gene was isolated from Fagopyrum esculentum Moench. (Fagopyrum esculentum Moench.) cultivar'Xinong 9976'by homologous cloning and RACE. The gene sequence was 1668 BP long and contained a 1374 BP open reading frame encoding 457 amino acids. Faes AP2 protein has two highly conserved AP2 (APETALA2) domains and a highly conserved nuclear localization signal region consisting of 10 amino acid residues at the front of the first AP2 domain. Phylogenetic analysis showed that Faes AP2 protein is closely related to Arabidopsis thaliana (L.) Heynh. AP2 transcription factor. The results showed that the gene was expressed in stamens and pistils of pin and thrum type flowers of buckwheat, but not in young leaves and perianth slices, and the expression level of the gene was significantly changed in different development stages of the two types of flowers, reaching the highest value in the rapid expansion period of anthers. Therefore, it was speculated that the gene might be involved in the development of buckwheat flowers. The regulation of floral organ development.
