基于基因芯片技術(shù)新疆漢族食管鱗癌差異表達(dá)mRNA的篩選

【摘要】:目的食管癌是全球常見的的惡性腫瘤,新疆為發(fā)病率較高的地區(qū)之一。本研究旨在完善新疆漢族食管鱗癌(esophageal squamous cell carcinoma,ESCC)基因表達(dá)譜,進(jìn)一步了解ESCC的發(fā)生、發(fā)展機(jī)制,提供與ESCC的預(yù)防、診斷及治療相關(guān)的新的生物標(biāo)志。方法收集2014-11-03-2015-09-25新疆石河子大學(xué)醫(yī)學(xué)院第一附屬醫(yī)院兵團(tuán)內(nèi)鏡中心6例漢族中分化ESCC患者的癌組織和癌旁正常組織(距癌組織5cm)。利用基因芯片技術(shù)檢測新疆漢族ESCC的差異表達(dá)基因。結(jié)果共篩選出漢族ESCC差異表達(dá)的mRNAs 335個,其中有138個表達(dá)上調(diào)的mRNAs和197個表達(dá)下調(diào)的mRNAs,差異倍數(shù)(fold change,FC)≥2且P0.01,這些基因涉及多種生物學(xué)功能及通路。結(jié)論利用基因芯片技術(shù)基于人類全基因譜篩選了新疆漢族ESCC差異表達(dá)基因,完善了新疆漢族ESCC基因譜,為ESCC的診斷及預(yù)后的進(jìn)一步研究打下基礎(chǔ)。
[Abstract]:Objective esophageal cancer is a common malignant tumor in the world. The purpose of this study was to improve the gene expression profile of (esophageal squamous cell carcinomatous carcinoma in Xinjiang Han nationality, to further understand the pathogenesis and mechanism of ESCC, and to provide a new biomarker related to the prevention, diagnosis and treatment of ESCC. Methods the cancer tissues and adjacent normal tissues (5cm) of 6 Han patients with differentiated ESCC were collected from the Endoscopic Center of Corps of the first affiliated Hospital of Shihezi University Medical College of Xinjiang on 2014-11-03-2015-09-25. The differentially expressed genes of ESCC in Xinjiang Han nationality were detected by gene chip technique.
