TRV介導(dǎo)的VIGS技術(shù)在果蔬基因功能研究中的應(yīng)用

【摘要】:病毒誘導(dǎo)的基因沉默(virus-induced gene silencing,VIGS)是近年來(lái)開(kāi)發(fā)的一種快速鑒定植物基因功能的反向遺傳學(xué)技術(shù)。因該技術(shù)需要借助病毒載體來(lái)實(shí)現(xiàn),所以病毒載體的開(kāi)發(fā)一直備受關(guān)注。在已開(kāi)發(fā)的病毒載體中,煙草脆裂病毒(Tobacco rattle virus,TRV)具有寄主范圍廣、沉默效率高、可在多個(gè)組織產(chǎn)生沉默,并且病毒誘導(dǎo)的癥狀輕微等獨(dú)特的優(yōu)勢(shì)。因此,TRV-VIGS技術(shù)已成功用于多種果蔬作物基因功能的研究。本文就TRV載體的結(jié)構(gòu)特點(diǎn)、作用機(jī)制、在果蔬基因功能研究中的應(yīng)用及存在的問(wèn)題等進(jìn)行了分析和總結(jié)。
[Abstract]:Virus-induced gene silencing (virus-induced gene silencing) is a rapid reverse genetic technique for the identification of plant gene function in recent years. Because the technology needs to be realized by virus vector, the development of virus vector has been paid more and more attention. Among the virus vectors developed, tobacco embrittlement virus (Tobacco rattle virustrv) has many unique advantages, such as wide host range, high silencing efficiency, silencing in multiple tissues and mild symptoms induced by the virus. Therefore, TRV-VIGS technology has been successfully used to study the gene function of many fruits and vegetables. In this paper, the structure and mechanism of TRV vector, its application in the study of gene function of fruits and vegetables and the existing problems are analyzed and summarized.
