廣州地區(qū)番石榴根結(jié)線蟲(chóng)鑒定與14-3-3基因的克隆

【摘要】:【目的】摸清廣州地區(qū)番石榴根結(jié)線蟲(chóng)的種類(lèi)、分布及致病機(jī)制!痉椒ā坑^察番石榴根結(jié)線蟲(chóng)的雄蟲(chóng)、二齡幼蟲(chóng)、雌蟲(chóng)及其會(huì)陰花紋的形態(tài)學(xué)特征,根據(jù)根結(jié)線蟲(chóng)的通用引物#C2F3和#1108對(duì)線蟲(chóng)mt DNA的COⅡ和lr RNA基因間序列進(jìn)行PCR擴(kuò)增,獲得750 bp的特異擴(kuò)增產(chǎn)物,將擴(kuò)增片段在Gen Bank上進(jìn)行Blast比對(duì)。通過(guò)南方根結(jié)線蟲(chóng)與北方根結(jié)線蟲(chóng)的14-3-3序列設(shè)計(jì)簡(jiǎn)并引物,進(jìn)一步擴(kuò)增已鑒定的象耳豆根結(jié)線蟲(chóng)的14-3-3基因!窘Y(jié)果】廣州地區(qū)番石榴根結(jié)線蟲(chóng)的形態(tài)與象耳豆根結(jié)線蟲(chóng)(Meloidogyne enterolobii)相似;其擴(kuò)增片段與象耳豆根結(jié)線蟲(chóng)(M.enterolobii)比對(duì),序列相似性在99%以上。擴(kuò)增了象耳豆根結(jié)線蟲(chóng)的14-3-3基因,結(jié)果顯示該基因的開(kāi)放閱讀框包含783 bp的片段,編碼261個(gè)氨基酸,命名為Me-14-3-3!窘Y(jié)論】廣州地區(qū)番石榴根結(jié)線蟲(chóng)為象耳豆根結(jié)線蟲(chóng)(M.enterolobii),14-3-3蛋白基因被成功克隆,擴(kuò)增結(jié)果為進(jìn)一步研究14-3-3蛋白基因在象耳豆根結(jié)線蟲(chóng)生長(zhǎng)發(fā)育中的功能、摸清致病機(jī)制等方面奠定良好的基礎(chǔ)。
