廣州地區(qū)番石榴根結(jié)線蟲鑒定與14-3-3基因的克隆

【摘要】:【目的】摸清廣州地區(qū)番石榴根結(jié)線蟲的種類、分布及致病機制!痉椒ā坑^察番石榴根結(jié)線蟲的雄蟲、二齡幼蟲、雌蟲及其會陰花紋的形態(tài)學(xué)特征,根據(jù)根結(jié)線蟲的通用引物#C2F3和#1108對線蟲mt DNA的COⅡ和lr RNA基因間序列進行PCR擴增,獲得750 bp的特異擴增產(chǎn)物,將擴增片段在Gen Bank上進行Blast比對。通過南方根結(jié)線蟲與北方根結(jié)線蟲的14-3-3序列設(shè)計簡并引物,進一步擴增已鑒定的象耳豆根結(jié)線蟲的14-3-3基因!窘Y(jié)果】廣州地區(qū)番石榴根結(jié)線蟲的形態(tài)與象耳豆根結(jié)線蟲(Meloidogyne enterolobii)相似;其擴增片段與象耳豆根結(jié)線蟲(M.enterolobii)比對,序列相似性在99%以上。擴增了象耳豆根結(jié)線蟲的14-3-3基因,結(jié)果顯示該基因的開放閱讀框包含783 bp的片段,編碼261個氨基酸,命名為Me-14-3-3!窘Y(jié)論】廣州地區(qū)番石榴根結(jié)線蟲為象耳豆根結(jié)線蟲(M.enterolobii),14-3-3蛋白基因被成功克隆,擴增結(jié)果為進一步研究14-3-3蛋白基因在象耳豆根結(jié)線蟲生長發(fā)育中的功能、摸清致病機制等方面奠定良好的基礎(chǔ)。
