【摘要】：按歐盟聯(lián)合研究中心(Joint Reaserch Center,JRC)的歐盟網(wǎng)絡(luò)轉(zhuǎn)基因?qū)嶒?yàn)室(European Network of GMO laboratories,ENGL)方法驗(yàn)證工作組編制的指南文件的要求,對《GB/T 19495.5—2004轉(zhuǎn)基因產(chǎn)品檢測核酸定量PCR檢測方法》進(jìn)行方法驗(yàn)證。分別驗(yàn)證了脫氧核糖核酸(deoxyribonucleic acid,DNA)提取質(zhì)量,方法的準(zhǔn)確度、重復(fù)性和相對定量限4個(gè)指標(biāo)。經(jīng)過驗(yàn)證,選用的DNA提取試劑盒提取質(zhì)量合格;檢測值在可接受參考值的±25%內(nèi),準(zhǔn)確度良好;相對可重復(fù)性標(biāo)準(zhǔn)偏差均小于25%,重復(fù)性良好;相對定量限為0.1%轉(zhuǎn)基因含量。依照指南對國標(biāo)進(jìn)行了驗(yàn)證,結(jié)果證明該指南在實(shí)驗(yàn)室進(jìn)行方法驗(yàn)證中具有指導(dǎo)性作用。
[Abstract]:According to the requirements of the European Network of GMO Laboratories (ENGL) method validation working group of the Joint Reaserch Center (JRC), the method validation of was carried out. The quality, accuracy, repeatability and relative quantitative limit of DNA extraction were 4 indexes. After verification, the quality of DNA extraction kit was qualified; the detection value was within (+) 25% of the acceptable reference value, the accuracy was good; the relative repeatability standard deviation was less than 25%, and the relative quantitative limit was good. The content of GM was 0.1%.
