柳蠶免疫基因Defense和Apolipophorin-Ⅲ的克隆與功能分析

【摘要】:昆蟲在外界不良生存環(huán)境下能夠有效地抵御外源病菌的入侵,除了機(jī)體保護(hù)色和物理屏障如鱗毛、甲殼和角質(zhì)表皮外,昆蟲也具有識別和除清體內(nèi)異原物的高效免疫系統(tǒng),用來抵御微生物的寄生和感染,因而其先天性免疫應(yīng)答機(jī)制的研究備受廣泛關(guān)注,這對闡釋生物的起源進(jìn)化、多樣性研究和生長發(fā)育有重要的意義。本研究以鱗翅目昆蟲柳蠶為研究對象,對柳蠶的defense基因和apolipophorin-Ⅲ(ApoLp-Ⅲ)基因進(jìn)行了克隆、鑒定、比較和進(jìn)化研究,也初步探討了它們的誘導(dǎo)調(diào)控模式和功能,以期豐富鱗翅目昆蟲先天免疫防御機(jī)制的理論基礎(chǔ),有利于我們了解新的昆蟲防御對策和病原物感染模式。本研究主要內(nèi)容與結(jié)果如下:1.從柳蠶中克隆并鑒定了defense基因,全長包含691個核苷酸預(yù)測的開放閱讀框498 bp,編碼165個氨基酸殘基并有一個卷曲結(jié)構(gòu)。defense蛋白的序列與其他鱗翅目昆蟲如蓖麻蠶、柞蠶、尺蠖、家蠶、美國白蛾、煙草天蛾等的defense蛋白序列有較高的同源性。在大腸桿菌中defense與表達(dá)載體PET-32a(+)連接并成功表達(dá)出包涵體和可溶性蛋白,并制備多克隆抗體用于后續(xù)實(shí)驗(yàn)。實(shí)時定量PCR和蛋白印跡檢測結(jié)果表明defense基因在脂肪體中有特別高的表達(dá)水平量,對柳蠶進(jìn)行免疫刺激后表明,defense基因可以被大腸桿菌、微黃藤球菌和核型多角體病毒刺激上調(diào)表達(dá),并表現(xiàn)出對真菌、病毒和革蘭氏陽性菌較強(qiáng)的敏感性,而對革蘭氏陰性菌的敏感性較弱。抑菌圈實(shí)驗(yàn)驗(yàn)證了抑菌圈直徑接近相同時,革蘭氏陽性菌所需的時間比陰性菌要短。這些數(shù)據(jù)說明,defense基因在柳蠶的先天免疫應(yīng)答中起著重要的作用。2.同時從柳蠶中克隆分離出ApoLp-Ⅲ基因,它包含660個核苷酸,預(yù)測的開放閱讀框561 bp并編碼186個氨基酸組成的蛋白,柳蠶ApoLp-Ⅲ蛋白序列與其他物種昆蟲的ApoLp-Ⅲ蛋白序列同源性很高,尤其是柞蠶。重組的ApoLp-Ⅲ質(zhì)粒在大腸桿菌中表達(dá)出可溶性的重組蛋白,并制備了多克隆抗體。qRT-PCR檢測柳蠶ApoLp-Ⅲ基因在脂肪體和表皮中表達(dá)量較高,而在血細(xì)胞、性腺、絲腺、馬氏管和中腸中很低。在抵御外源物如大腸桿菌、藤黃微球菌、球孢白僵菌和核型多角體病毒入侵時,ApoLp-Ⅲ基因均被上調(diào)表達(dá),并表現(xiàn)出對革蘭氏陰性菌和真菌比對革蘭氏陽性菌和病毒有更高的靈敏度。通過RNA干擾實(shí)驗(yàn),下調(diào)的ApoLp-Ⅲ基因直接或間接地影響著超氧化物歧化酶Cu/ZnSOD基因的表達(dá)也下調(diào),這些在免疫應(yīng)答反應(yīng)中起重要作用的相關(guān)基因之間可能存在著一定的相互作用的關(guān)系。
