偏頭痛模型大鼠降鈣素基因相關(guān)肽、腺苷A1、A2a受體的表達(dá)及天舒膠囊對(duì)其影響

【摘要】:目的研究降鈣素基因相關(guān)肽(calcitonin gene-related peptide,CGRP)、腺苷 A1、A2a受體(A1R、A2aR)在偏頭痛模型大鼠三叉神經(jīng)節(jié)(trigeminal ganglion,TG)和三叉神經(jīng)脊束尾核(spinal trigeminal nucleus caudalis, TNC)中的表達(dá)及天舒膠囊對(duì)其表達(dá)的影響。探討這三種物質(zhì)在偏頭痛大鼠模型中的表達(dá)特點(diǎn)和相互關(guān)系及天舒膠囊預(yù)防偏頭痛的可能作用機(jī)制。方法將40只大鼠隨機(jī)分為空白對(duì)照組(BC組,n=10)、假手術(shù)組(SO組,n=10)、電刺激三叉神經(jīng)節(jié)組(electrical stimulation of the trigeminal ganglion,ESTG 組,n=10)、天舒膠囊干預(yù)組(TC組,n=10)?瞻捉M處死前不做特殊處理,ESTG組電刺激三叉神經(jīng)節(jié)30min, SO組只插電針不給予電刺激,刺激時(shí)間同ESTG組,TC組大鼠進(jìn)行ESTG造模前按體質(zhì)量3.75 g/(kg·d)給予天舒膠囊灌胃,每天1次,持續(xù)7 d。最后1次灌胃后lh建立ESTG模型,模型復(fù)制方法同ESTG組。模型成功后,立即斷頭處死大鼠,快速取出大鼠的刺激側(cè)的TG及TNC所在的腦干部位,-80℃冰箱保存?zhèn)溆。采用REAL-TIME PCR、Western Blot技術(shù)分別檢測(cè)4組大鼠TG、TNC中CGRP、A1R、A2aR的表達(dá)量的變化。結(jié)果①在TG和TNC中,偏頭痛模型組大鼠的CGRPmRNA、A2aRmRNA的表達(dá)量高于空白對(duì)照組和假手術(shù)組(P0.05);偏頭痛模型組大鼠A1RmRNA表達(dá)量低于空白對(duì)照組和假手術(shù)組(P0.05)空白組和假手術(shù)組之間此三種物質(zhì)的表達(dá)量差異沒(méi)有統(tǒng)計(jì)學(xué)意義(P0.05)。②在TG和TNC中,偏頭痛模型組大鼠的CGRP、A2aR的蛋白表達(dá)量高于空白組和天舒膠囊干預(yù)組大鼠(P0.05), A1R的蛋白表達(dá)量低于空白組和天舒膠囊干預(yù)組(P0.05);空白組和天舒膠囊干預(yù)組之間此三種物質(zhì)的表達(dá)量差異沒(méi)有統(tǒng)計(jì)學(xué)意義(P0.05)。③CGRP、A2aR、A1R三種物質(zhì)之間的表達(dá)量的關(guān)系。在偏頭痛大鼠TG和TNC中,CGRP和A2aR兩者之間均存在正相關(guān)關(guān)系(P0.01);而CGRP和A1R兩者之間存在負(fù)相關(guān)關(guān)系(P0.01)。結(jié)論①CGRP、A2aR在偏頭痛大鼠模型中高表達(dá),A1R低表達(dá),參與偏頭痛的病理生理進(jìn)程。②預(yù)防應(yīng)用天舒膠囊影響CGRP、A2aR、A1R的表達(dá),對(duì)偏頭痛起到一定保護(hù)作用。③CGRP同A2aR、A1R之間存在相關(guān)關(guān)系,三種物質(zhì)之間相互作用參與偏頭痛的發(fā)病過(guò)程。
[Abstract]:Objective to study the expression of calcitonin gene-related peptide (calcitonin gene-related peptidea) and adenosine A1RtA2a receptor (A1R- A2aR) in trigeminal ganglion TG and (spinal trigeminal nucleus caudalis, TNC) of caudate nucleus of spinal tract of trigeminal nerve in migraine model rats and the effect of Tianshu capsule on the expression of CGRP. To investigate the expression and relationship of these three substances in migraine rat model and the possible mechanism of Tianshu capsule in preventing migraine. Methods Forty rats were randomly divided into three groups: blank control group (BC group), sham operation group (so group), (electrical stimulation of the trigeminal ganglion stimulation group (ESTG group) and Tianshu capsule intervention group (TC group). In the blank group, the trigeminal ganglion was stimulated 30 minutes without special treatment before death, the so group was treated with electroacupuncture only, and the stimulation time was the same as that in the ESTG group before the ESTG model was made. Tianshu capsule was given orally according to the body mass of 3.75 g / (kg d), once a day for 7 days. The ESTG model was established at lh after the last gastric perfusion, and the model was duplicated with the same method as that in the ESTG group. After the model was successfully established, the rats were killed immediately, and the stimulation side TG and the brain stem of TNC were quickly removed and stored in refrigerator at -80 鈩,
