多巴胺轉(zhuǎn)運體基因敲除小鼠認知功能的變化
[Abstract]:Objective To observe the changes in cognitive function of dopamine transporter (DAT) gene knockout mice. Methods Western blot and PCR method were used to detect the DAT knockout C57BL/6 J mouse model, and the changes in cognitive energy of mice were studied by Morris water maze test and open field test on DAT knockout mice. The results were in Morris water maze test and W. Compared with group T, the average escape latency of DAT gene knockout mice was obviously prolonged and its learning ability decreased. After the withdrawal of the platform, the retention time in the original platform quadrant shortened and its memory ability decreased. In the open field test, the total exploration path of the DAT knockout mice was lower than that of the WT mice, although the spontaneous excitement was independent. The activity of sexual movement increased, but most of it was a purposeful movement, and its retention time and times in the central lattice also decreased, indicating that the ability to explore curiosity was obviously decreased. Conclusion knockout mice DAT gene could lead to mice hyperactivity and damage the cognitive function of mice, indicating that the DAT gene may be related to learning, memory and exploration ability.
