巴西橡膠樹HbRPB11基因的克隆與表達(dá)分析

【摘要】:橡膠樹樹皮中的次生乳管是天然橡膠合成和貯存的主要場(chǎng)所。生產(chǎn)上施用乙烯利通過(guò)延長(zhǎng)橡膠樹割膠后的排膠持續(xù)時(shí)間,調(diào)節(jié)膠乳代謝,從而達(dá)到增產(chǎn)的目的。但目前對(duì)于轉(zhuǎn)錄復(fù)合體關(guān)鍵成員RNA聚合酶Ⅱ在膠乳代謝過(guò)程中的動(dòng)態(tài)變化尚不清楚。本研究采用RACE和RT-PCR方法,從橡膠樹無(wú)性系CATAS7-33-97的膠乳中克隆到編碼RNA聚合酶Ⅱ關(guān)鍵亞基RPB11的同源基因,命名為HbRPB11。該基因全長(zhǎng)659 bp,包含351 bp的開放閱讀框,編碼一個(gè)由116個(gè)氨基酸殘基組成的蛋白質(zhì)。該蛋白質(zhì)分子量為13.53 k D,理論等電點(diǎn)為5.934。實(shí)時(shí)熒光定量PCR結(jié)果表明,HbRPB11在橡膠樹的多個(gè)組織中均有表達(dá),其中在成熟葉片和膠乳中的表達(dá)量最高,并且受割膠和乙烯利處理顯著上調(diào)。橡膠樹不同種質(zhì)間的表達(dá)模式分析顯示HbRPB11基因在橡膠合成效率高的種質(zhì)中的表達(dá)量要顯著高于橡膠合成效率低的種質(zhì),表明該基因的表達(dá)量與橡膠合成效率呈正相關(guān)性,有可能作為生產(chǎn)上篩選高效合成橡膠種質(zhì)材料的分子標(biāo)記。
