產(chǎn)己酸菌Lysinibacillus fusiformis SigA基因的克

【摘要】:通過克隆產(chǎn)己酸菌Lysinibacillus fusiformis SigA全長基因,并對其進(jìn)行表達(dá)和生物信息學(xué)分析。結(jié)果表明,SigA基因開放閱讀框(ORF)有1 128個(gè)堿基,編碼327個(gè)氨基酸,其分子質(zhì)量為45.275 kDa;生物信息學(xué)預(yù)測表明,L.fusiformis與Lysinibacillus sphaericus具有較近的親緣關(guān)系,二者序列相似度達(dá)97%;該蛋白為親水性可溶性蛋白,與Bacillus sp.B14905相似度達(dá)100%,其二級(jí)結(jié)構(gòu)主要由α-螺旋和loop環(huán)狀構(gòu)成,α-螺旋和loop環(huán)狀分別占其二級(jí)結(jié)構(gòu)的60.15%和37.79%;預(yù)測并優(yōu)化的三級(jí)結(jié)構(gòu)由14個(gè)α-螺旋和15個(gè)loop構(gòu)成。
[Abstract]:The full-length Lysinibacillus fusiformis SigA gene of caproic acid producing bacteria was cloned and expressed and analyzed by bioinformatics. The results showed that the open reading frame (ORF) of SigA gene had 1,128 bases and encoded 327 amino acids, and its molecular weight was 45.275 kDa.The bioinformatics predicted that L.fusiformis and Lysinibacillus sphaericus had close relationship. The protein is hydrophilic soluble protein. Its secondary structure is mainly composed of 偽 -helix and loop ring, 偽 -helix and loop ring account for 60.15% and 37.79% of the secondary structure respectively, and the predicted and optimized tertiary structure is composed of 14 偽 -helix and 15 loop.
