限飼對湖羊子宮RGMb基因及BMP系統(tǒng)成員表達的影響
[Abstract]:The purpose of this study was to investigate the effects of feeding restriction on the expression of RGMb gene and BMP system members in uterus of Hu sheep. QPCR was used to detect the tissue expression profile of RGMb mRNA in 3 months old Hu sheep, and IHC was used to detect the localization of RGMb in uterus. After 35 days of pregnancy, Hu sheep were divided into two groups. The control group was fed with 100%NRC diet, the control group was fed with 50%NRC diet, and the uterine tissue (nb8) was collected on the 110th day of gestation. The expression of BMPR1A, BMPR1BU BMPR2 and RGMb in BMP2BMP4BMP4, BMPR1B, BMPR2 and RGMb were detected by qPCR and Western blotting (WB). Results RGMb mRNA was expressed in 14 kinds of tissues of Hu sheep, and the relative expression of RGMb mRNA in uterus was more than 0.5. The expression of RGMb protein in uterine epithelium and glandular epithelium of Hu sheep was significantly lower than that in control group (P0.05), but the level of mRNA had no significant change (P0.05), and BMP2 and BMP4 mRNA in the uterus of (R) Hu sheep were not significantly changed (P0.05), and the expression of RGMb protein in the uterus of (R) Hu sheep was significantly lower than that of control group (P0.05). Compared with the control group, the level of (C) increased significantly (P0.05), while the BMP receptor BMPR1 Agna BMPR1B and BMPR2mRNA levels decreased significantly compared with the control group (P0.05). The effects of feeding restriction on the expression of BMP protein, especially RGMb protein in uterine tissues, suggested that RGMb might be involved in the regulation of uterine homeostasis by BMP system in Hu sheep, which provided the experimental basis for further study on the mechanism of BMP in uterus.
