斜帶石斑魚生長(zhǎng)激素基因重組蛋白的發(fā)酵

【摘要】:將含有石斑魚生長(zhǎng)激素基因的畢赤酵母X33進(jìn)行震蕩培養(yǎng)后,裝入≥50L的自控罐中,28~30.5℃、pH 5.5~6.0、溶解氧≥30mg/L條件下發(fā)酵84h,生產(chǎn)石斑魚生長(zhǎng)激素基因重組蛋白。發(fā)酵期間,通過(guò)甲醇誘導(dǎo)畢赤酵母獲得目的蛋白的高效表達(dá),最后通過(guò)Western Blot和SDS-PAGE聚丙酰胺凝膠電泳對(duì)目的蛋白進(jìn)行定性定量檢測(cè)。最終獲得目的蛋白表達(dá)量約為1.5~2.0g/L,約占胞外可溶性總蛋白的16.4%。本試驗(yàn)用自控罐發(fā)酵生產(chǎn)含有斜帶石斑魚生長(zhǎng)激素基因重組蛋白的畢赤酵母生產(chǎn)工藝,為實(shí)現(xiàn)工業(yè)化生產(chǎn)魚類生長(zhǎng)激素餌料添加劑奠定基礎(chǔ)。
