木豆抗旱相關(guān)基因CcGST1克隆與表達(dá)分析

【摘要】:為探討谷胱甘肽轉(zhuǎn)移酶(GST)的抗逆機(jī)理,克隆木豆中的抗旱谷胱甘肽轉(zhuǎn)移酶基因,基于木豆干旱脅迫c DNA文庫(kù)中1個(gè)GST基因相關(guān)的EST片段,利用RACE技術(shù)從木豆幼苗中克隆了該基因,命名為CcGST1,研究其在不同組織及不同干旱時(shí)期的表達(dá)特性。結(jié)果表明:該基因含1個(gè)669 bp的開(kāi)放閱讀框,編碼223個(gè)氨基酸殘基,含有保守的谷胱甘肽-S-轉(zhuǎn)移酶的N端和C端結(jié)構(gòu)域;系統(tǒng)進(jìn)化樹(shù)分析結(jié)果表明木豆CcGST1與大豆、寬葉菜豆等豆科植物的GST親緣關(guān)系較近;亞細(xì)胞定位預(yù)測(cè)CcGST1蛋白可能定位于葉綠體;熒光定量PCR分析表明,CcGST1在木豆幼苗根、莖和葉中均有表達(dá),但根中的表達(dá)水平最高,同時(shí)該基因受干旱脅迫誘導(dǎo)表達(dá)。據(jù)此推測(cè)木豆CcGST1基因可能與木豆應(yīng)激干旱脅迫相關(guān)。
[Abstract]:In order to investigate the mechanism of stress resistance of glutathione transferase (GST), the drought-resistant glutathione transferase (DGST) gene was cloned from xylem bean, based on an EST fragment associated with GST gene in c DNA library under drought stress. The gene was cloned from woody bean seedlings by race technique and named CcGST1. The expression characteristics of CcGST1 in different tissues and different drought periods were studied. The results showed that the gene contained an open reading frame of 669 BP, encoding 223 amino acid residues and containing conserved glutathione--Stransferase N-terminal and C-terminal domains. CcGST1 protein may be located in chloroplast by subcellular localization. Fluorescence quantitative PCR analysis showed that CcGST1 was expressed in root, stem and leaf of woody bean seedlings, but the highest expression level was found in root. At the same time, the gene was induced to express under drought stress. It is suggested that the CcGST1 gene may be related to stress drought stress.
