轉(zhuǎn)基因檢測微流控恒溫擴(kuò)增芯片的研制

【摘要】:近年來環(huán)介導(dǎo)等溫擴(kuò)增技術(shù)(Loop-Mediated Isothermal Amplification,LAMP)越來越多地應(yīng)用于轉(zhuǎn)基因農(nóng)作物、微生物等的基因檢測中。為了提高相關(guān)檢測機(jī)構(gòu)的轉(zhuǎn)基因產(chǎn)品檢測效率,加快進(jìn)出口食品農(nóng)產(chǎn)品的通關(guān)速度,本文首次使用碟式芯片環(huán)介導(dǎo)恒溫擴(kuò)增技術(shù),在65℃恒溫條件下,對樣品中的P-Ca MV35S、T-NOS、NPTⅡ、BAR、PAT、Cry1Ac、EPSPS I、EPSPS II、FMV35S和GM-HRA等轉(zhuǎn)基因靶標(biāo)進(jìn)行同步檢測,檢出限可達(dá)到0.5%(m/m),實(shí)現(xiàn)一次上樣即可完成樣品的轉(zhuǎn)基因成分篩查。該方法特異性和靈敏度良好、檢測時間較短,可實(shí)時讀取擴(kuò)增結(jié)果。對實(shí)際樣品的檢測結(jié)果表明,該方法的檢測結(jié)果與實(shí)時熒光PCR方法的完全一致。本文開發(fā)的轉(zhuǎn)基因檢測微流控恒溫擴(kuò)增碟式芯片,可用于常見農(nóng)產(chǎn)品(大豆、玉米、大米和棉花等)的轉(zhuǎn)基因成分篩查,為轉(zhuǎn)基因產(chǎn)品的定性檢測提供了新的快速、靈敏、高通量技術(shù)平臺。
