文心蘭OnERS1全長(zhǎng)基因克隆及表達(dá)分析

【摘要】:為了了解乙烯信號(hào)受體在文心蘭切花衰老中的作用,利用RACE技術(shù)克隆了文心蘭乙烯受體基因OnERS1(Gen Bank登錄號(hào)為KP410729)。生物信息學(xué)分析表明,OnERS1基因編碼631個(gè)氨基酸,分子量為70 773.7,等電點(diǎn)為6.82,是跨膜蛋白,與其它植物的ETR乙烯受體蛋白的相似性在74%~99%之間,屬ETR1類(lèi)乙烯受體;虮磉_(dá)分析結(jié)果表明,OnERS1基因在根和蕊柱中表達(dá)量最高,在葉片、側(cè)瓣和花萼中次之,在莖和唇瓣中表達(dá)量最低;文心蘭切花衰老期間OnERS1基因的表達(dá)量在第0~6 d持續(xù)增加,第6 d后突然下降,第8 d后下降到一個(gè)極低的水平。與此同時(shí),花器官中的乙烯釋放從第8 d之前緩慢上升,第8~10 d急劇升高。OnERS1基因的時(shí)空表達(dá)特異性表達(dá)說(shuō)明它可能與花衰老有關(guān)。
[Abstract]:In order to understand the role of ethylene signal receptor in senescence, OnERS1 (GenBank accession number KP410729) was cloned by race technique. Bioinformatics analysis showed that OnERS1 gene encodes 631 amino acids with a molecular weight of 70773.7 and a isoelectric point of 6.82. It is a transmembrane protein. The similarity of ETR ethylene receptor protein with other plants is between 74% and 99%, belonging to ETR1 type ethylene receptor. The results of gene expression analysis showed that OnERS1 gene expression was the highest in root and column, followed by leaf, lateral valve and calyx, and the lowest in stem and lip, and the expression of OnERS1 gene continued to increase on the 6th day after senescence. On the 6th day, it suddenly decreased, and on the 8th day it dropped to a very low level. At the same time, ethylene release in flower organs increased slowly from the 8th day, and increased sharply on the 8th day. The spatio-temporal expression of OnERS1 gene indicated that it might be related to flower senescence.
