藥材甲熱激蛋白基因SpHsp60的克隆及溫度脅迫下的表達(dá)

【摘要】:本研究采用RT-PCR技術(shù)克隆了藥材甲熱激蛋白60(Heat shock protein 60,Hsp60)基因的cDNA全長(zhǎng)序列,命名為SpHsp60(GenBank登錄號(hào):KX778623).氨基酸序列分析表明SpHsp60具有Hsp60基因家族高度保守的基序,實(shí)時(shí)定量PCR技術(shù)檢測(cè)了不同溫度脅迫后該基因的表達(dá)量,結(jié)果表明:-5℃和0℃低溫處理2h,藥材甲成蟲體內(nèi)的SpHsp60的表達(dá)量均顯著高于對(duì)照組,且42℃高溫處理2h后的表達(dá)量也顯著高于對(duì)照組.說(shuō)明SpHSP60與藥材甲應(yīng)對(duì)極端溫度脅迫相關(guān).
[Abstract]:In this study, the full-length cDNA sequence of heat shock protein 60 Hsp60 gene was cloned by RT-PCR and named SpHsp60 (GenBank accession number: KX778623). Amino acid sequence analysis showed that SpHsp60 had a highly conserved motif of Hsp60 gene family. The expression of SpHsp60 gene was detected by real-time quantitative PCR under different temperature stress. The results showed that the expression of SpHsp60 was significantly higher than that of the control group at the temperature of 5 鈩,
