【摘要】:MYB基因家族在植物生長(zhǎng)發(fā)育和應(yīng)答逆境脅迫中有重要作用.通過(guò)RT-PCR和RACE技術(shù)從馬尾松中克隆獲得了PmMYB169全長(zhǎng)cDNA序列,分析了其在低磷脅迫下的表達(dá)特性.PmMYB169全長(zhǎng)為1 407bp,開(kāi)放閱讀框?yàn)?27bp,編碼308個(gè)氨基酸.生物信息學(xué)分析表明,PmMYB169蛋白序列中存在MYB家族結(jié)構(gòu)區(qū)域,屬于R2R3類(lèi)MYB轉(zhuǎn)錄因子;同源性分析表明,PmMYB169在MYB家族保守區(qū)域上同源性較高,它與北美云杉親緣關(guān)系最近.熒光定量PCR對(duì)低磷脅迫下PmMYB169的表達(dá)分析結(jié)果表明,PmMYB169的表達(dá)量隨低磷脅迫時(shí)間延長(zhǎng)呈上調(diào)趨勢(shì),說(shuō)明PmMYB169參與了低磷脅迫應(yīng)答;對(duì)不同組織的表達(dá)分析發(fā)現(xiàn),PmMYB169為組成型表達(dá),在馬尾松根莖葉中均有表達(dá),以葉的表達(dá)量最高.
[Abstract]:MYB gene family plays an important role in plant growth and response to stress. The full-length cDNA sequence of PmMYB169 was cloned from Pinus massoniana by RT-PCR and race. The expression characteristics of PmMYB169 under low phosphorus stress were analyzed. The full length of PmMYB169 was 1 407 BP, and the open reading frame was 927 BP, encoding 308 amino acids. Bioinformatics analysis showed that there was MYB family structure region in PmMYB169 protein sequence, which belonged to R2R3 MYB transcription factor, and the homology analysis showed that PmMYB169 had high homology in the conservative region of MYB family, and had the closest relationship with Picea spruce. The expression analysis of PmMYB169 under low phosphorus stress by fluorescence quantitative PCR showed that the expression of PmMYB169 increased with the prolongation of the time of low phosphorus stress, indicating that PmMYB169 was involved in the response to low phosphorus stress, and the expression of PmMYB169 was expressed in different tissues. They were expressed in roots and leaves of Pinus massoniana, and the highest expression was found in leaves.
