【摘要】：sHsp21(小熱激蛋白)是小熱激蛋白家族的成員,在外界環(huán)境壓力下能夠被誘導,熱激后能夠調(diào)節(jié)蛋白質(zhì)的折疊和保護細胞免受外界脅迫。本文研究了入侵雜草空心蓮子草Alternanthera philoxeroides生防天敵蓮草直胸跳甲的一個關鍵基因sHsp21,并對其進行熱激誘導表達以及RACE全長擴增。獲得全長cDNA 693 bp并完整的開放閱讀框567 bp,共編碼188個氨基酸。以30℃為對照,分別測定蓮草直胸跳甲Agasicles hygrophila SelmanVogt長沙種群和海南種群在模擬夏季特定10∶00-14∶00時段36℃、39℃高溫條件下sHsp21的表達量。連續(xù)熱激6 d,每天4 h熱激后取樣,實時熒光定量PCR分析顯示:湖南種群高溫39℃和36℃誘導下sHsp21表達量都顯著高于對照組30℃;海南種群36℃誘導下的表達量與對照組30℃差異不顯著,39℃誘導下的表達量顯著高于對照組30℃和36℃。結(jié)果表明,sHsp21在湖南和海南兩地理種群不同耐熱能力上起到重要作用,并且sHsp21的高表達可能對其蟲體產(chǎn)生副作用,為下一步深入探究其生物學功能奠定了一定的基礎。
[Abstract]:SHsp21 (small heat shock protein) is a member of small heat shock protein family, which can be induced under environmental pressure. Heat shock can regulate protein folding and protect cells from external stress. In this paper, a key gene, sHsp21, which is a key gene for the biocontrol of Alternanthera philoxeroides, a natural enemy, was studied, and its expression was induced by heat shock and the full-length amplification of race was carried out.
