【摘要】：為明確豬巨細(xì)胞病毒福建株(PCMV-FJ01)的gB基因特征,根據(jù)其序列特征,設(shè)計(jì)特異性引物對(duì)其進(jìn)行分段擴(kuò)增,并對(duì)目的片段進(jìn)行克隆測(cè)序。結(jié)果發(fā)現(xiàn),PCMV-FJ01株gB基因全長(zhǎng)為2 580bp,編碼859個(gè)氨基酸;其和GenBank數(shù)據(jù)庫(kù)中的PCMV分離株的gB基因的核苷酸和氨基酸同源性分別均在98.3%和97.9%以上。此外,本研究還發(fā)現(xiàn)部分(約50%)PCMV代表株的gB基因存在ACA三核苷酸缺失(436位氨基酸存在谷氨酰胺Q的缺失)。除ZZ株外,PCMV不同分離株的gB基因是否存在基因缺失和其遺傳進(jìn)化正相關(guān)。
[Abstract]:In order to identify the GB gene characteristics of porcine cytomegalovirus strain PCMV-FJ01, a specific primer was designed to amplify PCMV-FJ01, and the target fragment was cloned and sequenced. The results showed that the total length of GB gene of PCMV-FJ01 strain was 2580bp, encoding 859 amino acids, and the nucleotide and amino acid homology of GB gene was 98.3% and 97.9% respectively with PCMV isolated in GenBank database. In addition, some of the GB gene of PCMV were found to have deletion of ACA trinucleotide and amino acid at 436 amino acids, and the deletion of glutamine Q was found in some of the GB genes of PCMV. There was a positive correlation between gene deletion and genetic evolution of GB gene in different PCMV isolates except ZZ strain.
