日本鰻鱺IFN-γ基因的鑒定、表達(dá)模式及啟動子活性分析

【摘要】:為揭示魚類IFN-γ的生物學(xué)功能,研究從日本鰻鱺(Anguilla japonica)中克隆獲得了IFN-γ基因,命名為Aj IFN-γ。Aj IFN-γ具有脊椎動物IFN-γ的典型特征:包括4外顯子/3內(nèi)含子的基因結(jié)構(gòu)、C端的IFN-γ特征性氨基酸基序和1個核定位信號,以及6個α-螺旋反向平行構(gòu)成的二級結(jié)構(gòu)。Aj IFN-γ在日本鰻鱺所有組織中均低水平轉(zhuǎn)錄表達(dá),其中肝臟中表達(dá)量最高,其次是皮膚和頭腎。Poly I:C刺激和遲緩愛德華氏菌感染均可顯著誘導(dǎo)Aj IFN-γ在鰓、頭腎、體腎和(或)脾臟中的轉(zhuǎn)錄表達(dá),表明Aj IFN-γ能夠參與日本鰻鱺抗菌和抗病毒的免疫過程。此外,研究還克隆了Aj IFN-γ基因的5′調(diào)控區(qū)序列共1536 bp,并構(gòu)建了一系列Aj IFN-γ5′調(diào)控區(qū)刪節(jié)突變體,分析其啟動子活性,結(jié)果表明,上游 240/+136區(qū)域中含有起始Aj IFN-γ轉(zhuǎn)錄的關(guān)鍵啟動子調(diào)控元件, 1062/ 814區(qū)域存在轉(zhuǎn)錄的正調(diào)控元件,而 1252/ 1062區(qū)域存在轉(zhuǎn)錄的負(fù)調(diào)控元件。上述結(jié)果進一步豐富了魚類IFN-γ的基礎(chǔ)知識。
[Abstract]:In order to reveal the biological function of fish IFN- 緯, IFN- 緯 gene was cloned from Anguilla japonica of Japanese eel. Named AjIFN- 緯. AjIFN- 緯 has typical characteristics of vertebrate IFN- 緯: IFN- 緯 characteristic amino acid motif of C-terminal and a nuclear localization signal, including the gene structure of exon 4 / 3 intron. The expression of AjIFN- 緯 in all tissues of Japanese eel was lower than that in the liver, and the secondary structure composed of 6 偽 -helix reverse parallelism was the highest, and the expression of AjIFN- 緯 was the highest in the liver of Japanese eel. Secondly, the transcription expression of AjIFN- 緯 in Gill, head kidney, body kidney and / or spleen was significantly induced by skin and head-kidney. Poly I: C stimulation and Edouard tardy infection, indicating that Aj IFN- 緯 can participate in the anti-bacterial and anti-virus immune process of Japanese eel (Anguilla japonica). In addition, the 5'regulatory region of AjIFN- 緯 gene was cloned into 1536 BP, and a series of abridged mutants of AjIFN- 緯 5 'regulatory region were constructed, and the promoter activity was analyzed. There are key promoter regulatory elements for initiation of AjIFN- 緯 transcription in the upstream region of 240 / 136, positive regulatory elements for transcription in the 1062 / 1062 region, and negative regulatory elements for transcription in the 1252 / 1062 region. These results further enrich the basic knowledge of fish IFN- 緯.
