拜城油雞肌肉生長抑制素基因多態(tài)性與生產(chǎn)性能的相關(guān)性研究

【摘要】:試驗旨在研究肌肉生長抑制素(myostatin,MSTN)基因在拜城油雞中的多態(tài)性及其與生產(chǎn)性能的關(guān)系,探尋可用于拜城油雞選育的分子遺傳標(biāo)記。選取96只6周齡的拜城油雞公雞,隨機(jī)分為3組,每組32只,分別在12、18和24周齡時對其生產(chǎn)性能指標(biāo)進(jìn)行測定,同時采用PCR-RFLP方法檢測MSTN基因的多態(tài)性,分析MSTN基因多態(tài)性與拜城油雞生產(chǎn)性能的關(guān)系。結(jié)果顯示,在拜城油雞公雞中MSTN基因存在多態(tài)性,檢測到3種基因型:AA、GA、GG,GA為優(yōu)勢基因型,等位基因A為優(yōu)勢等位基因,不符合哈代-溫伯格平衡定律(P0.05)。在12周齡時,AA基因型個體的體重、屠體重、半凈膛重、全凈膛重、胸肌重和腿肌重均顯著高于GA基因型個體(P0.05);在18周齡時,GA基因型個體的體重、胸肌重和腿肌重均顯著高于GG基因型個體(P0.05);在24周齡時,AA基因型個體的體重、屠體重和半凈膛重均極顯著高于GG基因型個體(P0.01),全凈膛重和腿肌重顯著高于GG基因型個體(P0.05),GA基因型個體的體重、屠體重、半凈膛重和腿肌重均顯著高于GG基因型個體(P0.05),其余指標(biāo)在不同周齡不同基因型個體間差異不顯著(P0.05)。MSTN基因多態(tài)性與拜城油雞部分生產(chǎn)性能指標(biāo)密切相關(guān),推測該基因可以作為拜城油雞選育的分子遺傳標(biāo)記之一。
[Abstract]:The aim of the experiment was to study the polymorphism of myostatinine (MSTN) gene and its relationship with production performance in Baicheng oil chicken, and to explore the molecular genetic markers that can be used in breeding of Baicheng oil chicken. 96 6-week-old Beicheng fowls were randomly divided into 3 groups, 32 in each group, and their production performance indexes were measured at the age of 12 ~ 18 and 24 weeks respectively. The polymorphism of MSTN gene was detected by PCR-RFLP. To analyze the relationship between the polymorphism of MSTN gene and the performance of Baicheng fowl. The results showed that there was polymorphism of MSTN gene in Baicheng chicken cock. Three genotypes were detected as dominant genotype and allele A as dominant allele, which did not accord with Hardy-Weinberg equilibrium law (P0.05). At the age of 12 weeks, the body weight of AA genotype individuals, slaughtering weight, half bore weight, full weight, breast muscle weight and leg muscle weight were significantly higher than those of GA genotype individuals, and at 18 weeks old, the weight of GA genotype individuals was significantly higher than that of GA genotype individuals, and the weight of GA genotype individuals at 18 weeks of age was significantly higher than that of GA genotype individuals. The weight of pectoral muscle and leg muscle were significantly higher than those of GG genotype, and the weight of AA genotype was significantly higher than that of GG genotype at 24 weeks of age. The carcass weight and half bore weight of GG genotype individuals were significantly higher than those of GG genotype individuals (P 0.01), and the whole bore weight and leg muscle weight were significantly higher than those of GG genotype individuals with GG genotype P0.05 and GA genotype, and the slaughtering weight was significantly higher than that of GG genotype individuals. The weight of half bore and leg muscle were significantly higher than those of GG genotype individuals (P 0.05). There was no significant difference in other indexes among individuals of different genotypes at different weeks of age. The polymorphism of P0.05U. MSTN gene was closely related to some production performance indexes of Bicheng oil chickens. It is speculated that this gene can be used as one of the molecular genetic markers in the breeding of Baicheng oil chicken.
