【摘要】：顆粒結(jié)合型淀粉合酶(GBSSI)是植物直鏈淀粉合成的關(guān)鍵酶,主要由W_x基因編碼。本研究選用51份木薯種質(zhì)資源,設(shè)計(jì)引物對(duì)W_x基因進(jìn)行PCR擴(kuò)增后測(cè)序。通過(guò)對(duì)獲得的W_x基因序列進(jìn)行了遺傳多態(tài)性研究,表明其具有很大的保守性。分析W_x基因的SNP分布特點(diǎn)顯示其主要分布在第二內(nèi)含子和第四內(nèi)含子之間。通過(guò)生物軟件分析預(yù)測(cè)其SNP產(chǎn)生對(duì)顆粒結(jié)合型淀粉合酶(GBSSI)的物理化學(xué)、二級(jí)結(jié)構(gòu)和三維結(jié)構(gòu)的影響。本研究了解了不同種質(zhì)間W_x基因的差異,可為木薯的育種提供參考依據(jù)。
[Abstract]:Granular-bound starch synthase (GBSSI) is a key enzyme in amylose synthesis, which is mainly encoded by WSP x gene. In this study, 51 cassava germplasm resources were used to design primers for PCR amplification of WX gene and sequencing. The genetic polymorphisms of the Wapx gene were studied, and the results showed that it was highly conserved. The analysis of SNP distribution of WSX gene showed that it was mainly distributed between the second intron and the fourth intron. The effects of SNP production on the physicochemical secondary and three-dimensional structures of granular-bound starch synthase (GBSSI) were predicted by software analysis. In this study, the differences of WSP x gene among different germplasm were studied, which can provide reference for cassava breeding.
