湖羊和巴什拜羊FSHR基因多態(tài)性分析

【摘要】:本文旨在了解綿羊(湖羊和巴什拜羊)促卵泡激素受體(FSHR)基因3'-UTR特征,分析高、低繁殖力綿羊品種FSHR基因3'-UTR突變位點多態(tài)性差異,并探索其機(jī)制。研究獲得788 bp的湖羊和巴什拜羊FSHR基因3'-UTR序列,兩者一致性為98.17%,且均含有加尾信號、保守的ARE元件和多個miRNA結(jié)合位點;池DNA測序在FSHR基因終止密碼子后325nt處發(fā)現(xiàn)一個堿基T插入/缺失,命名為*325del T;多態(tài)性分析發(fā)現(xiàn)綿羊FSHR基因*325del T位點有3種基因型(TT型、T-型和--型),在湖羊群體中T為優(yōu)勢等位基因(頻率為0.604),而在巴什拜羊群體中-為優(yōu)勢等位基因(頻率為0.635);熒光素酶活性分析發(fā)現(xiàn)*325del T突變對綿羊FSHR轉(zhuǎn)錄活性無顯著影響。發(fā)現(xiàn)FSHR基因*325del T的多態(tài)性可能與綿羊繁殖性能有關(guān)。
[Abstract]:The aim of this study is to understand the 3'-UTR characteristics of the follicle stimulating hormone receptor (FSHR) gene of sheep (Hu sheep and basbai sheep) and to analyze the polymorphism of 3'-UTR mutation at the FSHR gene of high and low fecundity sheep, and to explore its mechanism. The study obtained the FSHR based 3'-UTR sequence of 788 BP in Hu sheep and bahshibai sheep, and the consistency is 98.17%, and both contain tails. Signals, conservative ARE elements and multiple miRNA binding sites; pool DNA sequencing after FSHR gene termination codon 325nt found a base T insertion / deletion, named *325del T; polymorphism analysis found that the *325del T loci of the ovine FSHR gene were 3 genotypes (TT type, type and type) in the lake sheep population (frequency of 0.60) 4), in the basbai sheep population - the dominant allele (frequency 0.635), the luciferase activity analysis found that the *325del T mutation had no significant effect on the FSHR transcriptional activity of sheep. It was found that the polymorphism of the FSHR gene *325del T may be related to the reproductive performance of sheep.
