海南五指山豬TLR2基因克隆及生物信息學(xué)分析

【摘要】:為了探討海南五指山豬TLR2基因的分子結(jié)構(gòu)特征及相關(guān)遺傳變異,為研究TLR2基因遺傳變異與免疫疾病的關(guān)系打下基礎(chǔ),試驗(yàn)采用PCR技術(shù)對(duì)該基因組序列進(jìn)行克隆、測(cè)序及相關(guān)生物信息學(xué)分析。結(jié)果表明:獲得2 649 bp的五指山豬TLR2基因的DNA序列,其基因編碼區(qū)全長(zhǎng)2 358 bp,編碼785個(gè)氨基酸;與Gen Bank中發(fā)布的普通豬TLR2基因序列相比,共發(fā)現(xiàn)9處突變,其中在編碼區(qū)序列存在2處突變,屬于有義突變;與普通豬、馬、牛、綿羊、人、黑猩猩、獼猴、犬、小鼠、雞、斑馬魚(yú)的氨基酸序列的一致性分別為99.7%、82.1%、81.4%、80.6%、78.4%、78.4%、77.8%、77.3%、68.2%、50.7%、40.4%;五指山豬TLR2編碼的蛋白主要位于質(zhì)膜,屬于分泌性蛋白、親水性蛋白和跨膜蛋白,有信號(hào)肽和5個(gè)糖基化修飾位點(diǎn),存在較多的絲氨酸(Ser)和蘇氨酸(Thr)磷酸化位點(diǎn);不同物種TLR2基因在進(jìn)化過(guò)程中具有較高的保守性。說(shuō)明TLR2基因可作為今后研究海南五指山豬免疫疾病的重要指標(biāo)之一。
[Abstract]:In order to study the molecular structure and related genetic variation of TLR2 gene in Wuzhishan pig, Hainan, and to study the relationship between genetic variation of TLR2 gene and immune disease, the genomic sequence was cloned by PCR technique. Sequencing and bioinformatics analysis. The results showed that the DNA sequence of the TLR2 gene of Wuzhishan pig was 2 649 BP, and its coding region was 2 358 BP, encoding 785 amino acids, and 9 mutations were found compared with the normal TLR2 gene sequence published in Gen Bank. There are 2 mutations in the coding region, which belong to the sense mutation; and ordinary pigs, horses, cattle, sheep, human beings, chimpanzees, macaques, dogs, mice, chickens, The amino acid sequences of zebrafish were 99.72.1and 81.4, respectively. The amino acid sequences of zebrafish were 78.4 and 78.4, respectively. The protein encoded by Wuzhishan pig TLR2 was mainly located in the plasma membrane, which belonged to secreted protein, hydrophilic protein and transmembrane protein, and had signal peptides and five glycosylation modification sites. There were more serine and threonine Thr-phosphorylation sites, and TLR2 genes of different species were highly conserved in the course of evolution. The results suggest that TLR2 gene can be used as one of the important indexes for the future study of immune diseases in Wuzhishan pigs in Hainan.
